摘要
为了提高无核葡萄胚挽救中幼胚的成苗率,对以红宝石无核葡萄为母本,巨玫瑰、玫瑰香和希姆劳特葡萄为父本的杂交胚珠进行胚挽救,分析不同父本、取样时间和培养基对杂交胚珠胚挽救效果的影响.结果表明,在胚的离体培养中,以巨玫瑰葡萄作父本更有利于胚的发育和萌发,红宝石无核×巨玫瑰、红宝石无核×希姆劳特和红宝石无核×玫瑰香杂交胚珠的最佳取样时期均为授粉后61 d,相应的胚萌发率分别为20.55%、6.56%和9.60%.分析不同发育培养基和萌发培养基对胚挽救效果的影响,适合红宝石无核×希姆劳特和红宝石无核×玫瑰香杂交胚发育的为N1培养基(Nitsch+1.5 mol/L IAA+0.5 mol/L 6-BA+0.5 mol/L GA3+60 g/L蔗糖+5 g/L琼脂+1 g/L活性炭,pH值为5.8),胚发育率分别为8.00%和13.92%.适合胚萌发成苗的为W1培养基(WPM+0.2 mg/L 6-BA+30 g/L蔗糖+5 g/L琼脂+1.5 g/L活性炭,pH值为5.8).通过根尖染色体倍性鉴定,确定红宝石无核×巨玫瑰的9株杂交幼苗中,4株为三倍体幼苗,5株为二倍体幼苗.
In order to enhance the seedling rate of embryo in the embryo rescue of seedless grape,the embryo rescue was conducted on the hybrid embryo,in which Red Ruby Seedless grape was adopted as female parent,and Jumeigui,Muscat Hamburg and Himrod Seedless were adopted as male parents respectively.The influence of different male parents,sampling dates and medium on the embryo rescue was analyzed.The results showed that among the investigated culture of seedless grape,in cross combinations with Red Ruby Seedless grape,Jumeigui grape as male parent is the most suitable for embryo development and germination.The optimal sampling dates were 61 days after bloom in Red Ruby Seedless×Jumeigui,Red Ruby Seedless×Muscat Hamburg and Red Ruby Seedless×Himrod Seedless,the germination rates of embryos of which were 20.55%,6.56%and 7.65%respectively.According to the analysis on the influence of different developed medium and germination medium types on the embryo rescue,N1 midium(Nitsch+1.5 mol/L IAA+0.5 mol/L 6-BA+0.5 mol/L GA3+60 g/L sucrose+5 g/L agar+1 g/L activated carbon,pH=5.8)was suitable for the development of hybrid embryo of Red Ruby Seedless×Himrod Seedless and Red Ruby Seedless×Muscat Hamburg,the germination rates of which were 8.00%and 13.95%respectively.W1 medium(WPM+0.2 mg/L 6-BA+30 g/L sucrose+5 g/L agar+1.5 g/L activated carbon,pH=5.8)was suitable for the embryo germination and seedling of the above two cross combinations.By the chromosome ploidy identification in root tip,four traploid plants and five diploid plants were obtained in Red Ruby seedless×Jumeigui.
