乳铁蛋白抑制常见口腔及肠道致病菌的体外活性研究

摘要 试验旨在研究乳铁蛋白对常见口腔/肠道致病菌的抑制和细菌生物膜清除作用。选择重组人乳铁蛋白(rhLF)、牛乳铁蛋白(bLF)和蛋清溶菌酶(阳性对照组),对金黄色葡萄球菌等7种菌进行抑菌、细菌生物膜形成和清除等试验。结果表明:(1)最低抑菌浓度试验表明,rhLF、bLF对金黄色葡萄球菌、嗜酸乳杆菌、沙门氏菌、牙龈卟啉单胞菌等具有较强的抑制作用;(2)抑菌圈试验表明,rhLF对金黄色葡萄球菌、嗜酸乳杆菌的抑制效果优于bLF,而对沙门氏菌、牙龈卟啉单胞菌的抑制效果差于bLF(P<0.05);(3)生物膜形成抑制试验表明,不同浓度的rhLF对金黄色葡萄球菌的抑制作用均显著高于bLF(P<0.05),浓度为5 mg/mL的rhLF抑制效果达到99%;(4)生物膜清除试验表明,不同浓度的rhLF对金黄色葡萄球菌的生物膜清除效果均显著高于bLF(P<0.05),浓度为5 mg/mL的rhLF抑制效果达到56%。研究结果表明,乳铁蛋白对常见口腔/肠道致病菌,尤其对金黄色葡萄球菌、嗜酸乳杆菌、沙门氏菌、牙龈卟啉单胞菌等,具有一定的抑制作用;同时对细菌生物膜形成具有抑制和清除作用。 The purpose of this research was to explore the inhibitory effect on common oral and intestinal pathogenic bacteria and bacterial biofilm removal of Lactoferrin.Recombinant human Lactoferrin(rhLF)from transgenic cattle,Lactoferrin(bLF)and Lysozyme(positive control group)were selected for bacteriostasis test,bacterial biofilm formation and bacterial biofilm clearance by the bacteria such as StapHylococcus aureus and so on.The results are as follow:(1)At the lowest inhibitory concentration test,rhLF and bLF had strong inhibitory effect on Staphylococcus aureus,Lactobacillus acidophilus,Salmonella and Porphyromonas gingivalis.(2)The inhibition effect of rhLF on Staphylococcus aureus and Lactobacillus acidophilus was better than that of bLF,while that on Salmonella and Porphyromonas gingivalis was lower than that of bLF(P<0.05).(3)The inhibition test of biofilm formation of Staphylococcus aureus showed that the rhLF of different concentrations was significantly higher than that of bLF(P<0.05),and the rhLF inhibition effect of 5mg/mL reached 99%.(4)The biofilm clearance test showed that different concentrations rhLF was significantly higher than that of bLF(P<0.05)on Staphylococcus aureus biofilm,and the rhLF inhibition effect of 5mg/mL reached 56%.Lactoferrin has a certain inhibitory effect on common oral intestinal pathogenic bacteria,especially Staphylococcus aureus,Lactobacillus acidophilus,Salmonella,Porphyromonas gingivalis,etc.At the same time,it can inhibit and clear bacterial biofilm formation.
