梓醇对AAPH损伤条件下施万细胞的影响

Effects of catalpol on schwann cells under AAPH injury conditions

摘要 目的:观察梓醇对AAPH损伤条件下的大鼠施万细胞96(RSC96)抗凋亡的影响。方法:通过体外培养RSC96,建立AAPH损伤时间浓度梯度模型:0、40、80、120、160、200mmol/L AAPH对RSC96损伤2、3、4h,选取合适的损伤浓度和时间,给予不同梓醇浓度0、5、10、50、100、150μmol/L进行干预,选取最佳的浓度。光镜观察RSC96细胞形态结构,CCK-8检测细胞活性程度。结果:AAPH损伤时间浓度模型显示浓度为120mmol/L,时间为3h为最佳浓度和时间,经不同浓度梓醇干预后,10μmol/L为最佳梓醇干预浓度。与正常对照组比较,模型组CCK-8吸光度OD值显著降低(P<0.05),细胞突起显著缩短。药物干预组和模型组比较,梓醇能显著提高细胞存活率与贴壁细胞数(P<0.05),吸光度OD值显著增加(P<0.05),细胞突起长度显著增长。结论:梓醇能减轻AAPH对RSC96的损伤,增加RSC96的细胞存活率,然其作用机制有待进一步探索。 Objective:To observe the effects of catalpol on rat schwann cell 96(RSC96)anti-apoptosis under AAPH injury.Methods:RSC96 was cultured in vitro,to establish the model of AAPH injury time and concentration gradient,different the concentration of AAPH of 0,40,80,120,160,200mmol/L damaging RSC96 by the different AAPH injury time of 2,3,4h.Selecting the right time and concentration,use different concentration catalpol of 0,5,10,50,100,150μmol/L to intervene RSC96.To select the best concentration,observe RSC96 cell morphology,CCK-8 used to detect the cell activity.Results:AAPH damage model showed that the concentration of 120mmol/L,time of 3h was optimal concentration and time.By different concentration of catalpol after the intervention,the group of concentration of 10μmol/L was the best,compared with blank control group,damage model group,CCK-8 absorbance OD value significantly reduced(P<0.05),the cell bulge shortened significantly.Intervention of catalpol and damage model group by comparison,catalpol can significantly increase the survival rate and the number of adherent cells(P<0.05),the OD value increasing significantly(P<0.05),bulge growing significantly.Conclusion:Catalpol can reduce the damage of AAPH to RSC96 and increase the cell survival rate of RSC96,but its mechanism needs to be further explored.
