摘要
目的:探讨胰岛素减轻急性心肌梗死模型大鼠氧化应激损伤的作用及机制。方法:选取24只雄性健康SD大鼠,随机分为假手术组(Sham组)、急性心梗组(AMI组)和胰岛素处理组(Ins组),每组各8只。在建立冠脉结扎致急性心肌梗死模型6 h后,超声心动图测量大鼠左室收缩末期内径(LVESD)、左室舒张末期内径(LVEDD);检测血浆肌酸激酶(CK)、乳酸脱氢酶(LDH)、一氧化氮(NO)水平及心肌组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;实时定量PCR检测大鼠心肌组织中蛋白激酶B(Akt)和内皮型一氧化氮合酶(eNOS)mRNA表达。结果:与AMI组比较,胰岛素处理可明显改善大鼠血流动力学指标,降低LVEDD、LVESD,上调心肌组织中Akt、eNOS的mRNA表达水平,降低MDA水平,升高SOD活性(P<0.05)。结论:胰岛素可保护急性心肌梗死大鼠心肌,其作用可能与激活Akt/eNOS信号通路和抑制氧化应激相关。
Objective:To investigate the effect and mechanism of insulin in reducing oxidative stress injury in rats with acute myocardial infarction(AMI).Methods:24 male healthy SD rats were selected to establish the rat model of AMI induced by coronary artery ligation,and were randomly divided into Sham group,AMI group and Ins group,8 cases in each group.The left ventricular end systolic diameter(LVESD)and left ventricular end diastolic diameter(LVEDD)were measured by echocardiography 6 h after modeling.Serum levels of creatine kinase(CK),lactate dehydrogenase(LDH)and nitric oxide(NO)were detected.The content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)in rat myocardial tissue were detected.The mRNA expressions of protein kinase B(Akt)and endothelial nitric oxide synthase(eNOS)in myocardial tissue of rats were detected by real-time quantitative PCR.Results:Compared with AMI group,insulin treatment could significantly improve the hemodynamic indexes of rats,reduce LVEDD and LVESD,up-regulate the mRNA expression levels of Akt and eNOS in myocardial tissue,reduce MDA level and increase SOD activity(P<0.05).Conclusion:Insulin can protect the myocardium of rats with acute myocardial infarction,and its effects may be related to activation of the Akt/eNOS signaling pathway and inhibition of oxidative stress.
