摘要
目的:研究黄芪甲苷(Astragaloside IV,AS-IV)对扑热息痛诱导肝损伤小鼠炎症因子表达的影响。方法:ICR清洁级小鼠40只,随机分为4组:即空白对照组、模型组、低剂量组(AS-IV 20 mg/kg)组、高剂量线(AS-IV 40 mg/kg),采用扑热息痛诱导建立小鼠急性肝损伤模型,HE染色检测肝脏组织病理学变化,ELISA法检测血清肿瘤坏死因子α(Tumor necrosis factorα,TNF-α)、白细胞介素1β(Interleukin 1β,IL-1β)、白细胞介素6(Interleukin-6,IL-6)水平,比较各组间炎症因子表达的差异。结果:AS-IV低、高剂量组(20、40 mg/kg)均能显著减轻扑热息痛诱导的小鼠肝损伤程度,与模型组相比,AS-IV(20、40 mg/kg)组TNF-α、IL-1β、IL-6水平显著降低(P<0.01)。结论:AS-IV能显著降低扑热息痛所致肝损伤小鼠炎症因子的表达,对肝损伤可起到一定治疗作用。
Objective:To study the effect of Astragaloside IV(AS-IV)on the expression of inflammatory cytokines in acetaminophen-induced liver injury in mice.Methods:Forty ICR mice were randomly divided into four groups(blank control group,model group,AS-IV low dose group 20 mg/kg and AS-IV high dose 40 mg/kg group).Acute hepatic injury model in mice was induced by acetaminophen.Hematoxylin eosin(H&E)staining was used to detect the pathological changes of liver tissue.ELISA was used to detect serum tumor necrosis factorα(TNF-α),interleukin 1β(IL-1β)and interleukin-6(IL-6)levels the inflammatiry cytokines.Results:AS-IV(20 mg/kg,40 mg/kg)significantly reduced the degree of liver injury in mice induced by acetaminophencompared with the model group,in AS-IV(20,40 mg/kg)group TNF-α,IL-1β,and IL-6 levels were significantly reduced(P<0.01).Conclusion:This study demonstrated that AS-IV can significantly reduce the expression of inflammatory factors in mice with hepatic injury caused by acetaminophen and play a therapeutic role in liver injury.
