摘要
目的观察经典方剂百合固金汤对不同出血模型小鼠的出血时间和凝血时间的影响,并探讨其作用机制。方法昆明小鼠随机分为生理盐水对照组、阳性药对照组以及百合固金汤高、中、低剂量组。建立小鼠剪尾出血和肝脏出血2个模型,探究百合固金汤对不同出血模型的出血时间的影响。同时,测定凝血4项、血小板数目、组织型纤溶酶原激活性(t-PA)及其抑制剂(PAI-1)活性。结果出血模型实验表明,百合固金汤能够显著缩短小鼠剪尾以及肝物理损伤后的出血时间(P<0.01)。机制研究表明,百合固金汤缩短了小鼠的活化部分凝血酶原时间(APTT)(P<0.01),而对凝血酶时间(TT)及凝血酶原时间(PT)无明显影响(P>0.05)。百合固金汤干预显著升高了小鼠血浆中的血浆纤维蛋白原时间(FIB)(P<0.01)含量,以及血小板含量(P<0.01)。同时发现,百合固金汤可以影响纤维蛋白溶解系统,具有提高t-PA的活性的作用(P<0.05),并且能够降低PAI-1活性(P<0.05)。结论百合固金汤具有显著的止血作用,其止血作用机制与激活内源性凝血途径、激活纤溶系统,以及促进血小板聚集相关。
Objective:To study the effect of classic prescription Baihe Gujin Decoction(BGD)on bleeding time and coagulation time in mice with different bleeding models,and to explore its mechanism of action,so as to provide basis for its clinical efficacy in treating hemoptysis.Methods:Kunming mice were randomly divided into the blank control,positive control group and BGD groups of high dose,medium dose and low dose.Tail cutting hemorrhage and liver hemorrhage models were established to explore the effect of BGD on bleeding time of different bleeding models.Meanwhile,four items of coagulation,platelet count,t-PA and PAI-1 activity were measured.Results:The bleeding model experiment showed that BGD could significantly shorten the bleeding time after tail cutting and liver physical injury(P<0.01).Mechanism study showed that BGD shortened APTT of mice(P<0.01),but had no significant effect on TT and Pt(P>0.05).BGD significantly increased FIB(P<0.01)and platelet(P<0.01).Meanwhile,BGD affected the fibrinolysis system,increased the activity of t-PA(P>0.05),and reduced the activity of PAI-1(P>0.05).Conclusion:BGD has significant hemostatic effect,and its hemostatic mechanism is related to activating endogenous coagulation pathway and fibrinolytic system,and promoting platelet aggregation.
