摘要
目的建立并验证测定五味子醇甲含量的方法。方法依利特C18色谱柱(250mm×4.6mm,5μm),梯度洗脱,流动相为乙腈(A)-0.1%磷酸(B)(0~20min,15%A;20.01~24min,20%~25%A;24.01~26min,25%~30%A;26.01~30min,30%~35%A;30.01~240min,35%~40%A;40.01~45min,40%~48%A;45.01~53min,48%~53%A);柱温为40℃;流速1.0ml/min;检测波长348nm。结果五味子醇甲在0.10~3.00μg(r=0.9995)范围内进样量与峰面积呈良好的线性关系,其精密度、重复性、稳定性RSD分别为1.03%、1.24%、1.03%,回收率为97.39%,RSD为1.43%。结论所建立的测定方法准确、简便、具有良好的重复性,可用于参志颗粒的质量控制。
Objective To establish and Validate a method for determing contents of Gomisin A.Method HPLC method was employed on a EITRI C18 colum(250 mm×4.6 mm,5μm).The mobile phase for?Acetonitrile-Water(0.1%Phosphoric acid)with gradient elution(0~20 min,15%A;20.01~24 min,20%~25%A;24.01~26 min,25%~30%A;26.01~30 min,30%~35%A;30.01~240 min,35%~40%A;40.01~45 min,40%~48%A;45.01~53 min,48%~53%A).The colum temperature was 40℃.The UV detection wavelength for Gomisin A was 348 nm.The flow rate was 1.0 ml/min.Results The linera ranges of Gomisin A was 0.10~3.00μg(r=0.9995).The average recovery was 97.39%,RSD为1.43%.Conclusion The method is accurate,simple and has a well reprocuci bility,which can be used as principles of quantitative analysis for Shenzhi Granules.
