枳椇子含药血清对酒精性肝损伤的抗氧化作用 被引量:4

Effects of Hovenia dulcis Thunb-containing Serum on Anti-oxidation in LO2 Cells Injured by Ethanol

摘要 为探究枳椇子对急性酒精性肝损伤细胞的作用及其机制。首先,SD大鼠枳椇子水提液及生理盐水灌胃后心脏取血,制备枳椇子含药血清及空白血清。然后用酒精诱导人LO_2细胞构建酒精损伤细胞模型,并随机将细胞分为正常组、模型组、空白血清组和不同剂量含药血清组,采用MTT法检测细胞存活率;酶标法检测丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)的酶活力,及丙二醛(MDA)含量;WST-1法检测超氧化物歧化酶(SOD)活性,最后分析评估枳椇子对急性酒精性肝损伤的保护作用。结果显示,与空白血清组比较,5%和10%含药血清组肝细胞存活率显著升高(P<0.05);ALT和AST活性显著降低(P<0.05);MDA含量明显降低,SOD活性显著升高(P<0.05)。表明枳椇子对酒精诱导的肝细胞损伤有一定的保护作用,可能通过抗氧化作用的机制来实现其保护作用。 This study was conducted to investigate the effects of Hovenia dulcis Thunb-containing serum(HDT-containing serum)on anti-oxidation in LO2 cells injured by ethanol and the underlying mechanism.Firstly,total of 16 Sprague-Dawley(SD)rats were randomly divided into control group and drug group.HDT-containing sera were prepared 2 hours after the gastric perfusion at 7th day.Then the LO2 cells used in the experiment were divided into the following groups:normal group,model group,control group,and drug group.Cells were injured in all groups except the normal group.Cell viability was detected using methyl-tetrazolium(MTT)chromatometry,and the degree of cell damage were analyzed measuring the levels of alanine aminotransferase(ALT),aspartate aminotransaminase(AST),malondialdehyde(MDA),and superoxide dismutase(SOD).The results showed that compared with the model group,the 5%and 10%HDT-containing serum groups had significantly higher survival rates(P<0.05),and significantly reduced ALT and AST activity(P<0.05).All serum groups had significantly decreased MDA content and significantly increased SOD activity(P<0.05).The data suggests that pre-treatment with HDT-containing sera has anti-oxidation effect on the LO2 cells injured by ethanol.
