摘要
目的:探讨肝癌患者循环miR-203a-3p表达与SOCS1和SOCS3的关系及意义。方法:选取50例拟行手术治疗的原发性肝癌患者为研究组,术前留取血样,术中留取病理组织和癌旁组织;另取50例同期入院体检的健康志愿者为对照组,留取血样。采用荧光定量PCR实验测定血样miR-203a-3p表达水平;免疫蛋白印迹实验测定组织SOCS1和SOCS3表达;甲基化特异性PCR实验测定组织SOCS1和SOCS3的甲基化水平。结果:研究组循环miR-203a-3p水平低于对照组,差异具有统计学意义(P<0.05)。研究组病理组织SOCS1和SOCS3表达水平低于癌旁组织,病理组织SOCS1和SOCS3甲基化程度高于癌旁组织,差异具有统计学意义(P<0.05)。研究组循环miR-203a-3p水平与组织SOCS1和SOCS3表达呈正相关性(r=0.576和r=0.475,P<0.05),与组织SOCS1和SOCS3甲基化程度呈负相关性(r=-0.585和r=-0.579,P<0.05)。结论:miR-203a-3p可能对原发性肝癌的SOCS1、SOCS3甲基化修饰有影响,有望为其临床诊治提供新靶点。
Objective:To investigate the relationship between circulating miR-203a-3p expression level and SOCS1 and SOCS3 in liver cancer patients and its significance.Methods:Fifty patients with primary liver cancer to be treated by surgery were selected as the study group.Blood samples were collected before operation,and pathological tissues and adjacent healthy tissues were collected during operation.Another 50 healthy volunteers admitted for physical examination at the same time were taken as the control group,and blood samples were taken.The expression level of miR-203a-3p in blood samples was determined by fluorescence quantitative PCR.The expression of SOCS1 and SOCS3 was determined by Western blot assay.Methylation levels of SOCS1 and SOCS3 were measured by methylation specific PCR.Results:The level of circulating miR-203a-3p in the study group was lower than that in the control group,and the difference was statistically significant(P<0.05).The expression levels of SOCS1 and SOCS3 in the pathological tissues of the study group were lower than those in the adjacent healthy tissues,and the methylation levels of SOCS1 and SOCS3 in the pathological tissues were higher than those in the adjacent healthy tissues,with statistically significant differences(P<0.05).The level of circulating miR-203a-3p in the study group was positively correlated with the expression of SOCS1 and SOCS3 in tissues(r=0.576 and r=0.475,P<0.05),and negatively correlated with the methylation degree of SOCS1 and SOCS3 in tissues(r=-0.585 and r=-0.579,P<0.05).Conclusion:miR-203a-3p may play an important role in the methylated modification of SOCS1 and SOCS3 in the primary liver cancer,which is expected to provide new targets for its clinical diagnosis and treatment.
