轮状病毒患犬与健康犬肠黏膜通透性及肠道菌群的差异性分析 被引量:3

摘要 为研究轮状病毒感染对犬肠黏膜通透性及肠道菌群的影响,选取2018年10月1日至2018年12月30日期间,在江苏农牧科技职业学院教学宠物医院就诊的轮状病毒感染患犬17例,记为A组,并设10例健康体检犬为对照,记为B组。用ELISA试剂盒检测血清中二胺氧化酶(DAO)、D-乳酸及内毒素(ET)的表达,每组随机选取9例,分别采集粪便,通过Illumina Miseq PE300平台进行细菌16S rDNA测序。结果表明:①与B组相比,A组DAO、D-乳酸和ET水平分别升高28.11%、12.37%和17.71%(P<0.01)。②A、B2组样品中共有OTU数226个,A组OTU数比B组低12.39%,主成分分析(PCA)显示A、B2组样本显著分开,肠道菌群相似度差异较大,A组肠道菌群ACE指数、Chao 1指数显著低于B组(P<0.05)。③在门水平上,2组样本中拟杆菌门、厚壁菌门、变形菌门和梭杆菌门为优势菌群,与B组相比,拟杆菌门和变形菌门比例升高,厚壁菌门比例减少(P<0.05);在属水平上,A组拟杆菌属、肠杆菌属、埃希氏菌属、克雷伯氏菌属等菌属数量增加,而双歧杆菌属、瘤胃球菌属、梭菌属等菌属数量与B组相比显著减少(P<0.05)。这一研究提示轮状病毒感染后可引起犬肠黏膜通透性升高,改变肠道菌群结构,加剧病情发展。 To study the effects of rotavirus infection on intestinal mucosal permeability and intestinal flora of dogs, 17 samples of canine rotavirus disease were selected as group A in teaching pet hospital of Jiangsu Agri-animal Husbandy Vocational College from Oct.1 2018 to Dec.30 2018, while 10 healthy dogs were set as the control(group B). DAO, D-lactic acid and ET in serum were detected by ELISA. Bacterial 16 S rDNA sequencing was performed by Illumina Miseq PE300 platform in feces of 9 samples randomly selected in each group. The results showed that: ① Compared with group B, the levels of DAO, D-lactic acid and ET in group A were respectively increased by 28.11%, 12.37% and 17.71%(P<0.01). ② The two groups had 226 OTUs in common, and the numbers of OTUs in group A were lower than group B by 12.39%.Principal component analysis(PCA) showed that the samples of group A were significantly separated from group B, and the similarity of intestinal flora was significantly different between two groups. The ACE index and Chao 1 index of intestinal flora in group A were significantly lower than group B(P<0.05). ③ At the level of phylum, the dominant bacteria contained Bacteroidetes, Firmicutes, Proteobacteria and Fusobacteria in two groups. Compared with group B,the proportion of Bacteroidetes and Proteobacteria were increased, while Firmicutes was decreased(P<0.05). At the level of genera, the proportion of Bifidobacterium, Rumincoccus and Clostridium in group A were lower than that in group B, while the proportion of Bacteroides, Enterobacter, Escherichia and Klebsiella were higher than group B(P<0.05). In conclusion, canine rotavirus infection can increase intestinal permeability, change the structure of intestinal flora, and aggravate the development of illness.
