摘要
谷子离体再生困难且品种间差异较大,限制了其在品种改良和相关研究中的应用。试验以晋谷29号、晋谷61号、张杂谷10号、张杂谷16号、冀谷39号、冀谷41号等6个谷子品种的种子为外植体开展离体再生培养,研究不同种类和浓度植物生长调节剂配比对谷子种子愈伤组织诱导、植株再生及生根的影响,以期建立稳定高效的谷子离体再生体系;并通过主体间效应检验方差分析,探究不同谷子品种和不同培养基对谷子愈伤组织诱导和分化作用的主次关系。结果表明,晋谷29号、晋谷61号、冀谷39号、冀谷41号的愈伤组织诱导培养基以MS+2.0 mg/L 2,4-D+0.2 mg/L 6-BA为好,张杂谷10号和张杂谷16号的愈伤组织诱导培养基以MS+1.0 mg/L 2,4-D+0.2 mg/L 6-BA为好;晋谷29号、晋谷61号、张杂谷10号、张杂谷16号和冀谷41号的愈伤组织分化培养基均以MS+1.0 mg/L 6-BA+0.5 mg/L NAA为好,冀谷39号的愈伤组织分化培养基以MS+2.0 mg/L 6-BA+0.5 mg/L NAA为好;6个谷子品种在各自适宜的培养条件下,其种子萌发率、愈伤组织诱导率和分化率均呈现较大差异;不同品种谷子试管苗生根差异不大,在1/2 MS培养基上均可诱导生根,且根系健壮,适于移栽。筛选离体再生率高的基因型和适宜的培养基可有效提高谷子组织培养和遗传转化效率,研究结果可为遗传转化在谷子上的应用及开展谷子的相关研究工作提供一定的理论依据。
Millet in vitro regeneration is difficult and the differences between varieties are large,which limits its application in variety improvement and related research.In this experiment,the seeds of six millet varieties(Jingu 29,etc.)were used as explants to carry out in vitro regeneration culture.The effects of different kinds and concentrations of plant growth regulators on the callus induction and plant regeneration of millet seeds were studied to establish a stable and efficient in vitro regeneration system of millet,and the test results were analyzed by variance analysis of the intersubjective effect,and the primary and secondary relationships of different millet varieties and different medium on the callus induction rate of millet were explored.The results showed that MS+2.0 mg/L 2,4-D+0.2 mg/L 6-BA was the best medium for callus induction of Jingu 29,Jingu 61,Jigu 39 and Jigu 41,MS+1.0 mg/L 2,4-D+0.2 mg/L 6-BA was the best medium for callus induction of Zhangzagu 10 and Zhangzagu 16,MS+1.0 mg/L 6-BA+0.5 mg/L NAA was the best medium for callus differentiation of Jingu 29,Jingu 61,Zhangzagu 10,Zhangzagu 16 and Jigu 41,MS+2.0 mg/L 6-BA+0.5 mg/L NAA was the best medium for callus differentiation of Jigu 39,and the seed germination rate,callus induction rate and differentiation rate of six varieties of millet were significantly different under the appropriate culture conditions.There was no significant difference in rooting among the six kinds of millet plantlets.Rooting could be induced on 1/2 MS medium,and the roots was robust and suitable for transplanting.Screening genotypes with high regeneration rate in vitro and suitable culture medium can effectively improve the efficiency of tissue culture and genetic transformation of millet.This experiment will provide a theoretical basis for the application of genetic transformation in millet and the related research work of millet.
