绵羊TSHR基因表达及其多态性与季节性发情之间的关系 被引量:3

摘要 为了探究绵羊TSHR基因表达及其多态性与季节性发情之间的关系,本试验采用实时荧光定量PCR(real-time fluorescent quantitative PCR,qPCR)研究TSHR基因在常年发情小尾寒羊和季节性发情苏尼特羊的大脑、下丘脑和垂体组织中的表达情况,比较两个绵羊品种的3种组织中TSHR基因的表达差异。同时利用SequenomMassARRAY○R SNP技术检测2组绵羊(常年发情组:小尾寒羊、策勒黑羊和湖羊;季节性发情组:滩羊、苏尼特羊和草原型藏羊)TSHR基因SNP位点(g.89290286A>G和g.89355312G>A)的多态性,并与绵羊季节性发情性状进行关联分析。结果表明,无论在小尾寒羊还是苏尼特羊中TSHR基因下丘脑中表达量均极显著高于大脑和垂体(P<0.01),且苏尼特羊下丘脑中TSHR基因表达量极显著高于小尾寒羊(P<0.01)。绵羊TSHR基因g.89290286A>G和g.89355312G>A位点均存在AA、GA和GG共3种基因型,其中g.89290286A>G位点在小尾寒羊、策勒黑羊和草原型藏羊中表现为中度多态(0.25<PIC<0.5),在滩羊、湖羊和苏尼特羊中表现为低度多态(PIC<0.25),g.89355312G>A位点在6个绵羊品种中表现为中度多态(0.25<PIC<0.5)。TSHR基因上述2个位点的基因型频率在常年发情和季节性发情绵羊品种间差异均达到极显著水平(P<0.01)。综上,TSHR基因的表达可能与绵羊季节性发情有关,其g.89290286A>G和g.89355312G>A位点与绵羊季节性发情性状可能存在一定关联。 To explore the expression and polymorphism of the TSHR gene and its association with seasonal estrus in sheep,real-time fluorescent quantitative PCR(qPCR)was used to detect the expression of TSHR gene in cerebrum,hypothalamus and pituitary in year-round estrus of Small Tail Han sheep and seasonal estrus of Sunite sheep,and compare the expression difference of TSHR in different breeds of sheep.SequenomMassARRAY○R SNP was applied to detect polymorphism of the SNPs(g.89290286A>G andg.89355312G>A)locus of TSHR gene in two different groups(year-round estrus group:Small-tail Han sheep,Celer black sheep and Hu sheep;seasonal estrus group:Tan sheep,Sunite sheep and Prairie sheep).Then the association was analyzed between TSHR gene and the trait of seasonal estrus.The results showed that the expression of TSHR gene in the hypothalamus was significantly higher than in cerebrum and pituitary(P<0.01)in both of Small Tail Han sheep and Sunite sheep.And the expression of TSHR gene in Sunite sheep in hypothalamus was extremely higher than that in Small Tail Han sheep(P<0.01).Three genotypes(AA,GA and GG)were found at g.89290286A>G andg.89355312G>A locus of TSHR gene in sheep.Population genetic analysis showed that theg.89290286A>G locus was at moderate polymorphism(0.25<PIC<0.5)in Small Tail Han sheep,Celer Black Sheep and Prairie sheep,and at low polymorphism(PIC<0.25)in Tan sheep,Hu sheep and Sunite sheep,and the g.89355312G>A locus was at moderate polymorphism(0.25<PIC<0.5)in 6 breeds of sheep.The genotype frequencies of the two loci were extremely significant between year-round estrous and seasonal estrous sheep breeds(P<0.01).In summary,the expression of TSHR gene may be related to the seasonal estrus of sheep,and g.89290286A>G and g.89355312G>A loci of TSHR gene might have a certain association with seasonal estrus of sheep.
