天然牛磺酸抗肝纤维化作用机制研究 被引量:6

摘要 目的研究天然牛磺酸(natural taurine,NT)对肝纤维化大鼠的保护作用,进一步探讨NT抗肝纤维化作用机制。方法将75只SD大鼠随机平均分为对照组、模型组、NT不同剂量组(0.3、0.6、0.9 g/d)。对照组大鼠正常饲养;模型组与NT各组大鼠均予皮下注射40%四氯化碳(CCl4)橄榄油溶液,首次剂量5 mL,以后每次3 mL,2次/周,共注射6周;NT各组予NT灌胃治疗,正常组及对照组予等量生理盐水,连续6周。实验末,ELISA检测大鼠血清IL-1β、IL-6、IFN-γ水平以及肝组织TGF-β1、TGF-β3、SOD、MDA及GSH-Px含量;RT-PCR检测肝组织TGF-β1、TGF-β3 mRNA表达量。结果与模型组比较,NT可以有效降低纤维化大鼠血清中炎症因子IL-1β、IL-6以及肝组织MDA、TGF-β1含量;NT可以有效促进肝纤维化大鼠血清IFN-γ及肝组织SOD、GSH-Px、TGF-β3分泌;NT可以下调肝纤维化组织TGF-β1 mRNA表达,促进TGF-β3 mRNA表达。结论 NT具有一定的抗肝纤维化作用,其机制可能与抑制大鼠肝脏的炎症反应、氧化应激作用以及下调TGF-β1 mRNA表达和促进TGF-β3 mRNA表达相关。 Objective To study the protective effect of natural taurine(NT) on hepatic fibrosis in rats and further explore the mechanism of NT in anti-hepatic fibrosis. Methods Seventy-five SD rats were randomly divided into control group, model group and NT different dose groups(0.3 g/d, 0.6 g/d, 0.9 g/d). Rats in the control group were fed normally. Rats in the model group and NT groups were subcutaneously injected with 40% carbon tetrachloride(CCl4) olive oil solution at the first dose of 5 mL, then 3 mL, 2 times/weeks for 6 weeks. Rats in NT groups were given NT intragastric administration, and rats in the normal group and the control group were given the same amount of saline for 6 weeks. At the end of the experiment, the levels of IL-1β,IL-6 and IFN-γ in serum and the contents of TGF-β1, TGF-β3, SOD, MDA and GSH-Px in liver tissue were detected by ELISA, and the expressions of TGF-β1 and TGF-β3 in liver tissue were detected by RT-PCR. Results Compared with the model group, NT could effectively reduce the levels of IL-1β, IL-6, MDA and TGF-β1 in serum, promote the secretion of IFN-γ in serum and SOD, GSH-Px and TGF-β3 in liver tissue of fibrotic rats, and down-regulate the expression of TGF-β1 and promote the expression of TGF-β3 in liver fibrotic tissues. Conclusion NT has a certain anti-fibrosis effect, and its mechanism may be related to inhibiting the inflammatory reaction of rat liver, oxidative stress, down-regulating the expression of TGF-β1 and promoting the expression of TGF-β3.
