摘要
目的构建心肌梗死大鼠模型,观察曲美他嗪对模型大鼠心肌细胞凋亡、自噬以及心功能的保护作用。方法60只SPF级成年雄性SD大鼠使用随机数字表法分为假手术组、对照组和曲美他嗪组。应用TUNEL法检测心肌细胞凋亡,并计算凋亡指数(PI),Western Blot检测心肌细胞自噬蛋白。彩超测定左心室收缩末期内径(LVESD)、短轴缩短分数(FS)和射血分数(EF)。结果假手术组、对照组、曲美他嗪组心肌细胞PI值分别为(0.76±0.23)%、(21.52±6.57)%和(12.38±2.15)%,心肌自噬蛋白LC3Ⅱ相对灰度值分别为0.17±0.03、0.46±0.12、0.68±0.15,3组间PI值和LC3Ⅱ相对灰度值比较差异有统计学意义(P<0.01)。术后第28天,假手术组LVESD、FS和EF与术前比较差异无统计学意义(P>0.05),对照组和曲美他嗪组LVESD较术前升高(P<0.05),FS和EF较术前减低(P<0.05)。曲美他嗪组LVESD术后第28天低于对照组(P<0.05),曲美他嗪组FS和EF术后第28天高于对照组(P<0.05)。结论曲美他嗪有助于心肌梗死大鼠心功能恢复,减低心肌细胞凋亡和增加心肌自噬。
Objective To observe the effects of trimetazidine on cardiac function,cardiomyocyte apoptosis and autophagy in rats with myocardial infarction.Methods Rat model of myocardial infarction was established.Sixty SPF-grade adult male Sprague-Dawley(SD)rats were randomly divided into sham-operated group,control group and trimetazidine group.TUNEL was used to detect cardiomyocyte apoptosis.Myocardial autophagy protein was detected by Western blot.Left ventricular end-systolic diameter(LVESD),fractional shortening(FS),and ejection fraction(EF)were detected by color Doppler ultrasound.Results The PI values of myocardial cells in sham operation group,control group and trimetazidine group were(0.76±0.23)%,(21.52±6.57)%,and(12.38±2.15)%,respectively.The relative gray values of myocardial autophagic protein LC3Ⅱwere(0.17±0.03),(0.46±0.12),and(0.68±0.15)in sham operation group,control group and trimetazidine group.There were significant differences in PI value and relative gray value of LC3Ⅱamong the three groups(P<0.01).At 28 days after the operation,LVEDD was significantly increased in the control group and trimetazidine group(P<0.05),while FS and EF were reduced significantly(P<0.05).The LVEDD value of the trimetazidine group was significantly lower than that of the control group at 28 days after surgery(P<0.05),and the FS and EF values of the trimetazidine group were significantly higher than those of the control group at 28 days after surgery(P<0.05).Conclusion Trimetazidine is helpful for the recovery of cardiac function in myocardial infarction rats,reduces myocardial cell apoptosis and increases myocardial autophagy,which may be its mechanism of action.
