摘要
目的评价姜黄人参提取物抗光老化的效果。方法以一定量的紫外线照射小鼠皮肤原代细胞和小鼠皮肤分别建立紫外线损伤的细胞、小鼠光老化模型;利用四唑盐比色法(MTT)和单细胞凝胶电泳法(SCGE)观察不同剂量的姜黄人参混合提取物对细胞损伤的保护作用,通过检测皮肤组织羟脯氨酸(HYP)、超氧化物歧化酶(SOD)和丙二醛(MDA)等含量评价姜黄人参混合提取物对小鼠皮肤光老化的抑制作用。结果与模型组相比,姜黄人参混合提取物能提高紫外线损伤后细胞的存活率、修复和保护损伤细胞DNA;降低小鼠皮肤组织MDA含量、提高HYP含量,增强SOD和谷胱甘肽氧化物酶(GSH-Px)活性(P<0.05)。结论姜黄人参提取物具有保护皮肤抗光老化的作用。
OBJECTIVE To evaluate the anti-photoaging effect of turmeric extract combined with ginseng extract.METHODS Ultraviolet light was used to induce lethal UV-injured model of mouse skin primary cells and photoaging model of mouse.The protective effect of turmeric extract and ginseng extract with different doses on cell damage in vitro was assessed by tetrazolium salt colorimetry(MTT)and single cell gel electrophoresis(SCGE).The effect of turmeric extract and ginseng extract with different doses on hydroxyproline(HYP),superoxide dismutase(SOD),malondialdehyde(MDA)were measured by Elisa.RESULTS The mixed extract of turmeric and ginseng could protect mouse skin primary cells against a lethal challenge with UV,as indicated by the increase of cell survival rate,repairing and protecting the DNA of damaged cells.In addition,the mixed extract of turmeric and ginseng could significantly reduce the MDA of mouse skin tissue,increase HYP content,enhance the activity of SOD and glutathione oxide enzyme(GSH-Px)to inhibit skin photoaging.CONCLUSION The mixed extract of turmeric and ginseng can protect skin against photoaging.
