紫草素通过TLR4/NF-κB信号通路抑制由LPS诱导的人牙髓细胞炎症研究 被引量:6

The effects of Shikonin on LPS-induced cell inflammation of human dental pulp cells through TLR4/NF-κB signaling pathway

摘要 目的:探讨紫草素对脂多糖(LPS)诱导的人牙髓细胞(HDPCs)细胞炎症的影响。方法:用MTT法检测不同浓度紫草素和LPS对HDPCs的细胞毒性;给药24 h后,通过MTT法检测细胞存活率;用ELISA法测定细胞液中IL-1β、IL-6、TNF-α和IL-10水平;通过Western blot法检测TLR4/NF-κB信号通路蛋白的表达情况。结果:紫草素可以抑制由LPS引起的细胞存活率的改变;与模型组相比,紫草素组中IL-1β、IL-6、TNF-α水平显著降低,IL-10水平显著升高;并且TLR4和p-NF-κB蛋白表达显著降低。结论:紫草素可以缓解LPS引起HDPCs细胞炎症,并且能够抑制TLR4/NF-κB信号通路蛋白的表达。 Objective:To investigate the effects of Shikonin on lipopolysaccharide(LPS)induced cell inflammation of human dental pulp cells(HDPCs).Methods:The cytotoxicity of Shikonin and LPS on HDPCs was detected by MTT assay.After treatment with different drugs for 24 h,MTT assay was applied to determine HDPCs cell viability.The secretion levels of IL-1β,IL-6,TNF-αand IL-10 were measured by ELISA.The protein expression of TLR4/NF-κB signaling pathway was determined by Western blot.Results:MTT results showed that Shikonin inhibited LPS induced change of HDPCs viability.Compared with the model group,Shikonin significantly reduced the secretion level of IL-1β,IL-6,TNF-αand increased the level of IL-10.Shikonin significantly decreased the protein expression of TLR4 and p-NF-κB.Conclusion:Shikonin can alleviate the cell inflammation of HDPCs induced by LPS and inhibit the protein expression of TLR4/NF-κB signaling pathway.
