摘要
【目的】研究尼罗罗非鱼(Oreochromis niloticus)干扰素诱导蛋白35(interferon induced protein 35,ifi35)在免疫反应中的作用。【方法】克隆获得罗非鱼ifi35基因开放阅读框序列(GenBank登录号:XM_003442606;On-ifi35),采用实时荧光定量PCR技术检测On-ifi35在健康罗非鱼不同组织中的分布,以及不同刺激物刺激后的表达。【结果与结论】On-ifi35基因编码区为1125 bp,编码374个氨基酸,理论分子质量为42.79 ku,等电点为5.01。On-ifi35含有1个LZD结构域和2个Nmi/IFP 35 domain(NID)。多序列比对发现On-ifi35与其它物种的ifi35氨基酸序列同源性介于35.23%~93.32%之间,且与斑马拟丽鱼同源性最高。系统进化分析发现,On-ifi35与斑马拟丽鱼的ifi35聚为一支。实时荧光定量PCR分析显示,On-ifi35在健康罗非鱼各组织中均有表达,其中在脾脏组织中表达量最高,其次是胸腺、肝脏、心脏、肌肉、肠道、体肾、脑、鳃、头肾,在皮肤中表达量最低。经无乳链球菌和PolyI:C刺激后,On-ifi35表达量在脾脏、肠道、体肾和头肾中均发生显著性变化且呈现出显著的时序性。
【Objective】To investigate the role of interferon induced protein 35(ifi 35)in the immune response of Oreochromis niloticus.【Method】The open reading frame of tilapia ifi 35 gene(GenBank accession number:XM_003442606;On-ifi 35)was cloned and the distribution of On-ifi 35 in different tissues of healthy tilapia and stimulation with different stimuli was detected by real-time fluorescent quantitative PCR.【Result and Conclusions】On-ifi 35 gene is 1125 bp,encoding 374 amino acids,the theoretical molecular mass is 42.79 ku,and the isoelectric point is 5.01.On-ifi 35 contains one LZD domain and two Nmi/IFP 35 domain(NID).Multiple sequence alignment revealed that On-ifi 35 shared 35.2%-93.3%sequence homology with ifi 35 from other species,and had the highest homology with Maylandia zebra.Phylogenetic analysis results indicate that On-ifi 35 was clustered with ifi 35 of the Maylandia zebra.Real-time quantitative PCR analysis showed that On-ifi 35 was expressed in all tissues of healthy tilapia.The highest expression was found in the spleen,followed by thymus,liver,heart,muscle,intestine,body kidney,brain,gill and head kidney.The lowest expression level was in the skin.After stimulation with Streptococcus agalactiae and PolyI:C,the expression level of On-ifi 35 was significantly changed in the spleen,intestine,mid-kidney and head kidney.
