摘要
缺氧诱导因子-1α(HIF-1α)是触发适应性反应的转录因子,在低氧条件下,对癌症代谢和治疗抗性方面起关键作用。HIF-1α的异常调节和过度表达已经与癌症生物学以及许多其他病理生理学有关,特别是在血管形成、能量代谢、细胞存活和肿瘤侵袭等领域。组织缺氧在子宫内膜癌生长过程中极为多见,HIF-1α作为关键转录因子,通过调控其下游基因的表达,在内膜癌缺氧耐受和恶行性进展方面发挥着重要的作用。近年来,靶向HIF-1α途径降低肿瘤进展和血管生成成为国内外研究的热点。众多研究发现,四硫钼酸盐、SHARP1、抗坏血酸、促红细胞生成素受体这4种物质,在机体内与其他细胞、生物因子、激素等共同发挥作用,通过不同的途径对HIF-1α的活性产生显著影响.为进一步探究HIF-1的生物活性及其相关的肿瘤治疗提供了更多的理论依据。
Hypoxia-inducible factor-1α(HIF-1α)is a transcription factor that triggers an adaptive response and plays a key role in cancer metabolism and therapeutic resistance under hypoxic conditions.Abnormal regulation and overexpression of HIF-1αhave been associated with cancer biology and many other pathophysiology,especially in angiogenesis,energy metabolism,cell survival,tumor invasion and other areas.Tissue hypoxia is very common in the growth of endometrial carcinoma.As a key transcription factor,HIF-1αplays an important role in hypoxia tolerance and malignant progression of endometrial carcinoma by regulating the expression of its downstream genes.In recent years,targeting HIF-1αpathway to reduce tumor progression and angiogenesis has become a hot topic.Many studies have found that tetrathiomolybdate,SHARP1,ascorbic acid,erythropoietin receptor,together with other cells,biological factors,hormones and so on,have significant effects on the activity of HIF-1αin different ways.It provides more theoretical basis for further exploring the biological activity of HIF-1 and its related tumor therapy.
