短期温度胁迫对西葫芦叶片抗坏血酸代谢系统的影响 被引量:3

Effects of short low and high temperature stress on ascorbic acid metabolism system in squash seedlings leaves

摘要 为了研究西葫芦叶片面对短期温度胁迫时抗坏血酸代谢系统的变化情况,以西葫芦‘德绿808’为试材,利用盆栽试验,通过光照培养箱设置低温(12℃/12℃,T1)和高温(38℃/38℃,T2)进行试验处理。结果表明,T1和T2胁迫显著增加了西葫芦幼苗叶片MDA含量、O2^ˉ·产生速率和H2O2含量,AsA、DHA和AsA+DHA含量,GSH、GSSG和GSH+GSSG含量,常温恢复后下降但仍高于对照。T1和T2处理下AsA/DHA和GSH/GSSG呈下降趋势,常温恢复后有回升但仍低于对照。T1和T2处理导致抗坏血酸过氧化物酶(APX)、单脱氢抗坏血酸还原酶(MDHAR)、脱氢抗坏血酸还原酶(DHAR)和谷胱甘肽还原酶(GR)活性明显升高,温度恢复后下降但仍高于对照。在整个胁迫期间,T2处理的以上指标均高于T1处理的,说明高温对西葫芦的伤害高于后者。以上结果说明了在温度胁迫初期,西葫芦叶片以 AsA为核心的抗氧化系统对抵御高温和低温胁迫发挥了重要作用 ,但是随着胁迫时间的延长,AsA代谢系统的抗氧化功能逐渐降低。 The study was to investigate the responses of ascorbic acid(AsA)metabolism system in squash leaves to low and high temperature stress,a pot experiment was conducted with squash plants(Delv 808)exposed to 12℃/12℃(T1)or 38℃/38℃(T2)treatment.The results showed that MDA contents,O2^-•producing rate and H2O2 content,AsA,DHA and AsA+DHA,GSH,GSSG and GSH+GSSG contents were significantly increased under T1 and T2 treatments,then decreased but higher than control after restoration under normal conditions.The ratios of ASA/DHA and GSH/GSSG were significantly decreased under T1 and T2 treatments,then increased but lower than control after restoration under normal conditions.The activities of ascorbate peroxidase(APX),monodehy droascorbate reductase(MDHAR),dehydroascorbate reductase(DHAR),and glutathione reductase(GR)under T1 and T1 stress were markedly increased,then decreased but higher than control after restoration under normal conditions.The changes of indexes under T1 treatments were higher than those under T1 treatments.All the results suggested that the AsA metabolism system in squash leaves was stimulated in the early period of high-and low temperature stresses,but its function declined gradually with the duration of the stresses.
