PBK/TOPK表达对膀胱癌BIU-87细胞株增殖及迁移的影响 被引量:4

The influence of PBK/TOPK expression on the proliferation and migration of BIU-87 bladder cancer cell line

摘要 目的探讨PBK/TOPK在膀胱癌中的表达及其表达对BIU-87细胞株增殖及迁移的影响。方法采用QRT-PCR及Western blot检测3例配对膀胱癌及癌旁正常组织中PBK/TOPK的表达。随后,对膀胱癌BIU-87细胞株中PBK/TOPK的表达分别干扰及过表达,通过CCK-8实验检测细胞的增殖,Transwell实验检测细胞的迁移能力。结果结果一致表明PBK/TOPK在膀胱癌中的表达明显高于癌旁正常组织(P<0.05)。干扰PBK/TOPK表达后,BIU-87细胞的增殖及迁移能力下降(P<0.05);而PBK/TOPK过表达后,BIU-87细胞的增殖及迁移能力增强(P<0.05)。结论PBK/TOPK高表达于膀胱癌,有助于膀胱癌细胞的增殖及迁移。 Objective To investigate the expression of PBK/TOPK and its influence on the proliferation and migration of BIU-87 bladder cancer cell line.Methods QRT-PCR and Western blot were used to detect PBK/TOPK expression in 3 cases of bladder cancer and matched normal tissues.Then,PBK/TOPK expression was knocked down and up-regulated in BIU-87 cell line.CCK-8 assay was used to assess cell proliferation,and Transwell assay was used to evaluate cell migration.Results QRT-PCR and Western blot results demonstrated that PBK/TOPK was highly expressed in bladder cancer tissues compared with those in normal tissues(P<0.05).Meanwhile,PBK/TOPK knockdown significantly inhibited cell proliferation and migration,but PBK/TOPK overexpression significantly promoted cell proliferation and migration(P<0.05).Conclusions PBK/TOPK is highly expressed in bladder cancer,which is contributed to cell proliferation and migration.
