摘要
目的考察当归补血汤中阿魏酸、毛蕊异黄酮、芒柄花黄素、黄芪甲苷在大鼠血浆中的药动学。方法UPLC⁃MS/MS法测定4种成分含有量,分析采用Waters BHE C18色谱柱(100 mm×2.1 mm,1.7μm);流动相0.1%甲酸⁃乙腈,梯度洗脱;体积流量0.4 mL/min;柱温30℃。大鼠灌胃给予药物冻干粉溶液,于不同时间点采血,DAS 3.0软件拟合血药浓度⁃时间曲线,计算药动学参数。结果阿魏酸、毛蕊异黄酮、芒柄花黄素、黄芪甲苷在各自范围内线性关系良好(r≥0.9951),提取回收率91.42%~124.83%。四者Cmax分别为312.30、799.39、149.19、120.46 ng/mL,Tmax分别为19.29、21.43、53.57、70.71 min,t1/2分别为192.37、173.95、185.34、408.77 min,AUC0~t分别为19220.97、111546.04、29073.89、18401.73μg/(L·min),AUC0~∞分别为20136.85、118807.61、34089.10、19921.64μg/(L·min)。结论该方法快速、灵敏、准确,可为当归补血汤临床应用和质量监控提供参考。
AIM To investigate the pharmacokinetics of ferulic acid,isoflavonein,formononetin and astragalo⁃side A in Danggui Buxue Decoction in rat plasma.METHODS UPLC⁃MS/MS was adopted in the content determi⁃nation of four constituents,the analysis was performed on a 30℃thermostatic Waters BHE C18 column(100 mm×2.1 mm,1.7μm),with the mobile phase comprising of 0.1%formic acid⁃acetonitrile flowing at 0.4 mL/min in a gradient elution manner.Rats were given intragastric administration of drug freeze⁃dried powder solution,after which blood collection was made at different time points,then blood concentration⁃time curve was fitted by DAS 3.0 software,and pharmacokinetic parameters was calculated.RESULTS Ferulic acid,isoflavonein,formonone⁃tin,astragaloside A showed good linear relationships within their own ranges(r>0.9951),whose extraction recov⁃eries were 91.42%-124.83%.These four constituents demonstrated the Cmax of 312.30,799.39,149.19,120.46 ng/mL,Tmax of 19.29,21.43,53.57,70.71 min,t1/2 of 192.37,173.95,185.34,408.77 min,AUC0-t of 19220.97,111546.04,29073.89,18401.73μg/(L·min),and AUC0-∞of 20136.85,118807.61,34089.10,19921.64μg/(L·min),respectively.CONCLUSION This rapid,sensitive and accu⁃rate method can provide references for the clinical application and quality monitoring of Danggui Buxue Decoction.
