摘要
目的:从水通道蛋白角度研究蚕沙的"化湿"作用。方法:SD大鼠,随机分为5组,即正常组、模型组、蚕沙低、中、高剂量组。采用"外湿侵体+正气耗损+过食肥甘"的方法,建立"湿阻中焦"证模型,Western blot法检测肾AQP1、肺AQP1、结肠AQP3、皮肤AQP3、颌下腺AQP5蛋白的表达,qPCR法检测AQP mRNA的表达。结果:与正常组比较,模型组大鼠肾中AQP1蛋白和mRNA表达明显升高(P<0.05),且肺AQP1、结肠AQP3、皮肤AQP3、颌下腺AQP5蛋白和mRNA表达均明显降低(P<0.05);与模型组比较,蚕沙高剂量组肾AQP1蛋白和mRNA表达显著降低(P<0.05),肺AQP1、结肠AQP3、皮肤AQP3、颌下腺AQP5蛋白和mRNA表达均显著升高(P<0.05)。结论:蚕沙可能通过调节肾、肺、结肠、皮肤、颌下腺组织中AQP的表达,发挥"化湿"的作用。
OBJECTIVE To study the"dampness-resolving"effect of Faeces Bombycis from the perspective of aquaporins.METHODS SD rats were randomly divided into five groups:the normal group,the model group,the low,middle and high dose groups of Faeces Bombycis.The rats were used to establish the syndrome of"damp retention in middle-Jiao"model by applying the method of"outer damp invades+righteous gas is exhausted+eating overfull fat and sweetmeat".The expression of protein and mRNA of AQP1 in renal and lung tissue,AQP3 in colon and skin tissue,AQP5 in submandibular gland tissue were detected by Western blot and qPCR.RESULTS Compared with the normal group,the expression of AQP1 protein and mRNA in renal tissue in the model group was significantly increased(P<0.05),the expression of protein and mRNA of AQP1 in lung tissue,AQP3 in colon and skin tissue,AQP5 in submandibular gland tissue in the model group were significantly decreased(P<0.05);compared with the model group,the expression of AQP1 protein and mRNA in renal tissue in the high dose group was significantly reduced(P<0.05),and the protein and mRNA expression of AQP1 in lung tissue,AQP3 in colon and skin tissue,AQP5 in submandibular gland tissue in the model group were significantly increased(P<0.05).CONCLUSION Faeces Bombycis may play a role of"dampness-resolving"by regulating the expression of AQP in renal,lung,colon,skin and submandibular gland tissue.
