摘要
海藻糖是生物体合成的、具有抗逆保护作用的天然二糖,在食品加工与贮藏、微生物发酵过程具有重要作用。该文采用前期获得的专一性海藻糖酶,建立了一种定量测定海藻糖的快速方法。该方法对海藻糖的检测限为0.019 mg/m L,定量限为0.1 mg/m L;海藻糖的线性检测范围为0.1 mg/m L^150.0 mg/m L,回收率为98.80%~101.33%。该方法操作简便快捷、特异性高、重复性好,在菌种改良与食品原料营养筛查等海藻糖快速定量检测中具有良好的应用前景。
Trehalose is a natural disaccharide synthesized by living organisms with stress tolerance and protection.It plays an important role in food processing and storage,as well as in microbial physiology.In this study,a rapid method for quantifying trehalose was developed using a selective trehalase which was obtained previously.The detection limit was 0.019 mg/m L and its quantification limit was 0.1 mg/m L.The linear range of detection was between 0.1 mg/m L and 150.0 mg/m L.The product recovery rate was 98.80%-101.33%.This new method is simple,rapid,with high specificity and reproducible.It has potential applications in quantifying trehalose during strain development and raw material nutrient analysis.
