摘要
目的探讨雌二醇化学发光免疫测定法中置换剂的影响和优化。方法采用竞争一步法原理建立E2化学发光免疫分析检测体系。对置换剂达那唑和甲二氢睾酮进行考察和优化,按照选定的两种置换剂的水平进行混合使用,对空白限、线性、特异性进行评估,并与进口全自动化学发光试剂检测结果进行对比。结果本系统线性范围为20~4 000pg/mL,最低检出限为5.1 pg/mL,与雌二醇类似物的交叉反应率均小于2%。与进口全自动化学发光试剂同时测定183份样本的E2水平,二者测定结果的相关系数r为0.9981,平均相对偏差为0.1%。结论评估了两种E2的置换剂,并优化了其使用水平。混合使用这两种置换剂可以达到理想的效果。混合使用置换剂的方法也为其他小分子的免疫诊断提供了一种置换剂使用的思路。
Objective To evaluate the influence and optimization of blocking agent in chemiluminescent immunoassay for Estradiol(E2).Methods The E2 chemiluminescence immunoassay detection system was established by the principle of competitive one-step method.Investigation and optimization of the blockers danazol and methyl dihydrotestosterone were conducted.The blank limit,linearity,and specificity were evaluated and compared with the results of imported fully automated chemiluminescence reagents.Results The linear range of the system was 20-4000 pg/mL,with the minimum detection limit 5.1 pg/mL,and the cross-reaction rate with estradiol analogs was less than 2%.The E2 concentration of 183 samples was measured simultaneously with the imported fully automatic chemiluminescence reagent.The correlation coefficient r of two determination results was 0.9981,and the average relative deviation was 1.2%.Conclusion Two blockers were evaluated and their working concentrations were optimized.Mixing these two blockers can achieve the desired blocking effect.The combination of blocking agents also provides a principle for the immunodiagnosis of other small molecules.
