促黄体素对绵羊卵母细胞体外成熟及卵裂的影响

摘要 促黄体素对卵母细胞成熟有一定的促进作用。为研究不同浓度LH(促黄体素)对绵羊卵母细胞体外成熟及受精发育的影响,从屠宰场采集绵羊卵巢,切割法获取卵母细胞,于不同浓度LH成熟培养液中培养24 h,进行体外受精及体外胚胎培养。按照LH的添加浓度将试验分为5个组(A组,0μg/mL;B组,5μg/mL;C组,10μg/mL;D组,15μg/mL;E组,20μg/mL)。试验结果表明:LH的添加浓度为10μg/mL时,卵母细胞的成熟率为76.60%,卵裂率为63.24%,桑椹胚率为30.81%,均显著高于其它试验组(P<0.05)。综上,本试验在含有10μg/mL FSH的TCM199成熟培养液中添加10μg/mL的LH时能显著提高绵羊卵母细胞的成熟率、卵裂率及桑椹胚率,这对建立稳定有效的绵羊卵母细胞成熟体外培养体系有着重要意义。 Luteinizing Hormone can promote oocyte maturation.In order to study the effect of different concentrations of LH(luteinizing hormone)on in vitro maturation and fertilization of ovine oocytes.Ovine ovaries were collected from slaughterhouse,and oocytes were obtained by the cutting method.After 24 hours of culture in different concentrations of LH mature medium,IVF and in vitro embryo culture were carried out.According to the added concentration of LH,the test was divided into 5 groups(group A,0μg/mL;group B,5μg/mL;group C,10μg/mL;group D,15μg/mL;group E,20μg/mL).The results showed that when LH concentration was 10μg/mL,the oocyte maturation rate was 76.60%,cleavage rate was 63.24%,and morula rate was 30.81%,which were significantly higher than other experimental groups(P<0.05).In conclusion,when 10μg/mL LH was added to TCM199 maturation medium containing 10μg/mL FSH,the maturation rate,cleavage rate and morula rate of ovine oocytes could be significantly increased,which is of great significance to establish stable and effective ovine oocyte maturation culture system in vitro.
