疏风解毒胶囊广谱抗病毒功效研究 被引量:33

摘要 目的:评价疏风解毒胶囊广谱抗病毒作用。方法:采用细胞病变(CPE)抑制法,以连花清瘟胶囊为对照,从药物的最大无毒浓度开始,测定疏风解毒胶囊对甲型流感病毒H1N1型(FM1株、PR8株、江西修水株、B10株、B59株)、单纯疱疹病毒1型、单纯疱疹病毒2型、呼吸道合胞病毒、副流感病毒(仙台株)、柯萨奇病毒B4型、柯萨奇病毒B5型以及腺病毒的抑制作用,按Reed-muench法计算50%细胞毒性浓度(TC50),最大无毒浓度(TC0),50%抑制浓度(IC50)和药物治疗指数(TI)。采用副流感病毒致小鼠肺炎模型,以利巴韦林颗粒和连花清瘟胶囊为对照,测定疏风解毒胶囊对模型小鼠肺指数的抑制作用。结果:疏风解毒胶囊的TC50为2.230g/L,TC0为1.560g/L;疏风解毒胶囊对甲型流感病毒H1N1型FM1株、PR8株、江西修水株、B10株、B59株的IC50均为0.56 g/L,TI均为3.98;疏风解毒胶囊对单纯疱疹病毒1型的IC50为0.79 g/L,TI为2.82;疏风解毒胶囊对单纯疱疹病毒2型的IC50为1.12g/L,TI为2.00;疏风解毒胶囊对呼吸道合胞病毒的IC50为0.32g/L,TI为7.00;疏风解毒胶囊对副流感病毒仙台株的IC50为0.14g/L,TI为15.93;疏风解毒胶囊对柯萨奇病毒B4型的IC50为0.56g/L,TI为3.98;疏风解毒胶囊对柯萨奇病毒B5型的IC50为0.28 g/L,TI为7.96;疏风解毒胶囊对腺病毒没有抑制作用。与模型组比较,疏风解毒胶囊低剂量、中剂量、高剂量对副流感病毒致小鼠肺炎模型肺指数均有抑制作用,但低剂量差异没有统计学意义,中剂量、高剂量有显著差异(P<0.01)。结论:疏风解毒胶囊在体外有广谱的抗病毒作用,对副流感病毒所致的小鼠肺炎也有较好的治疗作用。 Objective:To evaluate the broad-spectrum antiviral effect of Shufeng Jiedu capsules.Methods:Adopted the inhibition method of cytopathic effect(CPE)and took Lianhua Qingwen capsules as control,starting from the maximum of non-toxic concentration of drugs,and measured the inhibitory effect of Shufeng Jiedu capsules for influenza virus A of H1N1 type(FM1 strain,PR8 strain,Jiangxi Xiushui strain,B10 strain,B59 strain),herpes simplex virus type 1,herpes simplex virus type 2,respiratory syncytial virus,parainfluenza virus(Sendai strain),coxsackie virus type B4,coxsackie virus type B5 and adenovirus;calculated the cytotoxic concentration of 50%(TC50),the maximum of non-toxic concentration(TC0),the inhibition concentration of 50%(IC50)and the drug treatment indexes(TI)according to the Reed-muench method.Adopted the model of mouse pneumonia caused by parainfluenza virus and took ribavirin granules and Lianhua Qingwen capsules as control,and detected the inhibitory effect of Shufeng Jiedu capsules for the pulmonary indexes of model mice.Results:The TC50 of Shufeng Jiedu capsules was 2.230 g/L,and TC0 was 1.560 g/L;the IC50 of Shufeng Jiedu capsules for FM1 strain,PR8 strain,Jiangxi Xiushui strain,B10 strain,B59 strain of influenza virus A of H1N1 type was 0.56 g/L and the TI was 3.98;the IC50 of Shufeng Jiedu capsules for herpes simplex virus of type 1 was 0.79 g/L and the TI was2.82;the IC50 of Shufeng Jiedu capsules for herpes simplex virus of type 2 was 1.12 g/L and the TI was 2.00;the IC50 of Shufeng Jiedu capsules for respiratory syncytial virus was 0.32 g/L and the TI was 7.00;the IC50 of Shufeng Jiedu capsules for Sendai strain of parainfluenza virus was 0.14 g/L and the TI was 15.93;the IC50 of Shufeng Jiedu capsules for coxsackie virus type B4 was 0.56 g/L and the TI was 3.98;the IC50 of Shufeng Jiedu capsules for coxsackie virus type B5 was 0.28 g/L and the TI was 7.96;Shufeng Jiedu capsules has no inhibitory effect on adenovirus.Compared with the model group,all the low-dose,middle-dose and high-dose of Shufeng Jiedu capsules had inhibitory effects on the pulmonary indexes of the model of mouse pneumonia caused by parainfluenza virus,but there was no significant difference in the low-dose,and there was obvious difference in middle-dose and high-dose(P<0.01).Conclusion:The application of Shufeng Jiedu capsules in vitro has a broad-spectrum antiviral effect and has a good therapeutic effect on the mouse pneumonia caused by parainfluenza virus.
