摘要
目的:观察电针傍刺对褥疮大鼠创面组织中p38MAPK蛋白表达的影响。方法:入选的SD大鼠被分为正常对照组、模型对照组、模型抑制剂组、电针傍刺组和电针抑制剂组,又分为1天、3天、5天、7天、9天5个时间点,每个小组6只,共150只大鼠。各组褥疮大鼠创面均采用碘伏消毒,模型对照组腹腔注射生理盐水,电针傍刺组腹腔注射生理盐水后采用电针傍刺治疗,模型抑制剂组腹腔注射SB203580,电针抑制剂组腹腔注射SB203580后再采用电针傍刺治疗。分别用免疫印迹和免疫组化法检测各组大鼠创面组织中p38MAPK蛋白的表达。结果:模型对照组在1天和3天时p38MAP蛋白表达逐渐升高,在5天、7天和9天时逐渐下降。而电针傍刺组在1天时p38MAP蛋白表达升高,在3天、5天、7天和9天时逐渐下降。模型抑制剂组与模型对照组比较及电针抑制剂组与电针傍刺组比较p38MAPK蛋白表达下降。结论:电针傍刺能够促进p38MAPK蛋白的表达,SB203580能够抑制电针傍刺通过p38MAPK通路促进创面愈合的治疗作用。
Objective:To investigate the effect of electro-acupuncture(EA)Bangci(straight and side needling)on the expression of p38MAPK of wound tissue in rats with bedsore.Methods:The selected SD rats were divided into the normal group,the model group,the model inhibitor group,and EA Bangci group and EA inhibitor group.It is divided into five time points,i.e.1 d,3 d,5 d,7 d and 9 d.There were 6 rats in each group,and 150 rats in total.All the wounds of bedsore were treated with Iodophor;the model control group was intraperitoneally injected with normal saline;EA Bangci group was treated with EA Bangci after intraperitoneal injection of normal saline;the model inhibitor group was intraperitoneally injected with SB203580;the EA inhibitor group received intraperitoneal injection of SB203580 before EA Bangci.The expression of p38MAPK protein was detected by western blot and immunohistochemistry.Results:In the model group,p38MAP protein expression gradually increased on 1 d and 3 d,and gradually decreased on 5 d,7 d and 9 d.However,the expression of p38MAPK protein increased on 1 d in the EA Bangci group,and decreased gradually on 3 d,5 d,7 d and 9 d.The expression of p38MAPK protein decreased in the model inhibitor group compared to that in the model group,and it decreased in EA inhibitor group compared to that in EA group.Conclusion:Electro-acupuncture Bangci can promote p38MAPK protein expression,wheareas SB 203580 can inhibit the action to prevent the Wound healing.
