哺乳期过度营养及高脂膳食对大鼠胰腺脂肪酶蛋白水平的影响 被引量:1

EFFECTS OF LACTATION OVERNUTRITION AND HIGH-FAT DIET ON PANCREATIC LIPASE EXPRESSION IN RATS

摘要 目的探讨哺乳期过度营养及高脂膳食对胰腺脂肪酶的影响。方法本实验使用经典小窝模型模拟大鼠哺乳期过度喂养,通过调整乳鼠喂养只数分成哺乳期正常喂养组(NL组,10只/窝)和哺乳期过度喂养组(SL组3只/窝),并在出生3 w断乳后予以高脂膳食及正常饮食,形成哺乳期正常喂养组+断乳后正常摄食组(NL组),哺乳期过度喂养组+断乳后正常膳食组(SL组)、哺乳期正常喂养组+断乳后高脂膳食组(NL-HF组)和哺乳期过度喂养组+断乳后高脂膳食组(SL-HF组)。用自动化生化分析仪检测生化相关指标。化学方法萃取并测定大便中脂肪含量。用免疫印迹方法测定甘油三酯脂酶(pancreatic triglyceride lipase,PTL)与其相关蛋白2(pancreatic lipase-related protein 2,PLRP2)、胆盐刺激性脂酶(bile salt-stimulated lipase,BSSL)蛋白水平。结果与NL组大鼠比较,断乳后SL组大鼠体重增加(P<0.05),16 w时TG(Triglycerides)水平增加,糖耐量受损(P<0.05)。SL-HF组大鼠体重及TG水平持续高于其他各组(P<0.05),糖耐量受损;NL-HF组大鼠体重高于NL组(P<0.05),并于16 w出现TG升高和糖耐量受损(P<0.05)。NL-HF和SL-HF组大鼠胰腺PTL、PLRP2及BSSL的蛋白表达水平明显增高(P<0.05),但SL组与NL组胰腺脂肪酶PLT、BSSL、PLRP2蛋白表达无明显差异(P>0.05)。结论哺乳期过度喂养导致糖耐量受损,而断乳后的高脂膳食可诱导胰腺脂肪酶的表达增加,促进膳食脂肪的吸收。 Objective To explore the long term effect of the effects of lactation over-nutrition and high-fat diet on pancreatic triglyceride lipase expression.Methods A classical rat small litter model was used to simulate lactation over-nutrition in rats.Male Sprague-Dawley rat pup litters were adjusted to litter sizes of ten with normal diet(normal litters,NL,10 per litter)and three with lactation over-nutrition(small litters,SL,3 per litter).The groups were provided with high-fat diet or normal diet separately after three weeks lactation.Normal feeding group in the two periods was marked as NL.Lactation over-nutrition feeding with normal feeding after weaning group was marked as SL.Normal feeding before weaning and high-fat diet feeding after weaning group was marked as NL-HF while over-feeding before weaning combined with high-fat diet feeding after weaning group was marked as SL-HF.Automatic biochemical analyzer was used to detect relative indices.Chemical method was used to analyze the fat content in faeces.The expressions of PLT(pancreatic triglyceride lipase),PLRL2(ancreatic lipase-related protein 2)and BSSL(Bile Salt-Stimulated Lipase)were analyzed by Western blot.Results Compared with NL,body weight of the SL rats was higher(P<0.05)and at the end of the 16th week,SL had a increased level of TG and a decreased OGTT level(P<0.05).Rats in SL-HF group were heavier than any of the other groups(P<0.05)and had a decreased OGTT level.Rats in the NL-HF group had higher body weight than the NL group(P<0.05)and an increased level of TG and OGTT at the end of the 16th week(P<0.05).The expressions SL and NL of PTL,PLRP2 and BSSL were significantly increased(P<0.05)in rats of the NL-HF and SL-HF groups while in the SL and NL groups the expression showed no difference(P>0.05).Conclusion Over-feeding before weaning tends to influence the pancreatic endocrine while high-fat feeding after weaning leads to increased expression of pancreatic lipase and promoted fat absorption.
