甲状腺乳头状癌组织中FMNL2与Snail mRNA及蛋白表达的研究 被引量:1

Expression of FMNL2 and Snail genes in papillary thyroid carcinoma

摘要 目的探讨甲状腺乳头状癌(papillary thyroid carcinoma,PTC)组织中同源形成素样蛋白2(formin-like protein 2,FMNL2)、Snail mRNA及蛋白的表达水平及其临床意义。方法采用实时荧光定量PCR (FQ-PCR)及免疫组化En vision法分别检测2017年1月至2018年1月于台州市中心医院进行手术切除PTC样本60例和30例相应癌旁组织中FMNL2、Snail mRNA及蛋白的表达水平,采用Spearman等级相关分析FMNL2和Snail蛋白表达的相关性。结果 PTC组织中FMNL2和Snail mRNA表达量均高于癌旁组织的(P<0.05);PTC组织中FMNL2和Snail蛋白的阳性表达率分别为61.67%(37/60)和66.67%(40/60),均高于癌旁组织的13.33%(4/30)和10.00%(3/30)(P<0.05)。FMNL2 mRNA和蛋白表达与包膜侵犯(t=3.627,P=0.001;χ~2=11.487,P=0.001)、淋巴结转移(t=3.642,P=0.001;χ~2=5.958,P=0.014)及临床分期(t=3.228,P=0.002;χ~2=8.963,P=0.003)有关。Snail mRNA和蛋白在PTC的不同临床分期(t=2.914,P=0.005;χ~2=9.187,P=0.002)、包膜是否侵犯(t=3.683,P=0.000;χ~2=10.909,P=0.001)及淋巴结是否转移(t=3.595,P=0.001;χ~2=6.338,P=0.012)中的表达差异有统计学意义。PTC中FMNL2和Snail蛋白之间的表达呈正相关(r=0.533,P=0.000)。结论 FMNL2、Snail mRNA和蛋白表达升高可能在PTC的发生、包膜侵犯、淋巴结转移及临床分期进展的过程中发挥作用。FMNL2、Snail蛋白之间的表达明显的正相关,显示两者可能具有相似或协同的促癌作用。 Objective To observe the expression of formin-like protein 2(FMNL2) and Snail genes in papillary thyroid carcinoma(PTC) and to discuss its clinical significance. Methods 60 PTC tissue samples and 30 samples of corresponding adjacent tissues were collected from patients underwent surgeries in Taizhou Central Hospital from January 2017 to Janury 2018, the levels of FMNL2 and Snail mRNA as well as proteins were determined by real-time fluorescence quantitative PCR(FQ-PCR) and immunohistochemical en vision method.The correlation between the expression of FMNL2 and Snail genes was analyzed by Spearman rank correlation. Results The mRNA levels of FMNL2 and Snail in PTC tissues were significantly higher than those in adjacent tissues(P<0.05) with the positive expression rates of FMNL2 and Snail protein of 61.67%(37/60) and 66.67%(40/60), which were also significantly higher than those in adjacent tissue(P<0.05).The expression of FMNL2 was associated with capsule invasion(t=3.627, P=0.001;χ~2=11.487, P=0.001), lymph node metastasis(t=3.642,P=0.001;χ~2=5.958, P=0.014) and clinical stage(t=3.228,P=0.002;χ2=8.963,P=0.003).Meanwhile,the expression of Snail gene varied significantly in different clinical stages(t=2.914,P=0.005;χ2=9.187,P=0.002),capsule invasion(t=3.683,P=0.000;χ2=10.909,P=0.001)and lymph node metastasis(t=3.595,P=0.001;χ2=6.338,P=0.012).There was a positive correlation between FMNL2 and Snail gene expression in PTC(r=0.533,P<0.05). Conclusions The elevated levels of mRNA and protein of FMNL2 and Snail may play a certain role in the occurrence of PTC,envelope invasion,lymph node metastasis and disease progression.There is a significantly positive correlation between the expression of FMNL2 and Snail genes,suggesting that they may have similar or synergistic carcinogenic effects.
