摘要
目的:利用模式动物斑马鱼研究自噬对其尾鳍再生的影响。方法:选取成年斑马鱼剪去尾鳍,随后腹腔注射自噬抑制剂3-甲基腺嘌呤(3-Methyladenine,3-MA)或诱导剂STF-62247,连续注射2天,剪取再生尾鳍,通过qPCR分析自噬相关基因ATG和Beclin-1的表达。斑马鱼剪去尾鳍,连续7天腹腔注射3-MA或STF-62247,每天记录斑马鱼尾鳍再生长度。结果:尾鳍再生过程中,3-MA下调ATG和Beclin-1表达,STF-62247提高ATG表达,抑制自噬或诱导自噬对斑马鱼尾鳍再生速度无显著影响。结论:腹腔注射自噬抑制剂3-MA或自噬诱导剂STF-62247对斑马鱼尾鳍再生速度无显著影响。
Objective:To explore the effects of autophagy on zebrafish caudal fin regeneration.Methods:Adult zebrafish caudal fins amputated with were selected.Then,the adult zebrafish was injected with the autophagy inhibitor 3-Methyladenine(3-MA)or the activator STF-62247 once a day for two consecutive days.The regenerated caudal fin was collected to analyze the expressions of autophagy related genes ATG and Beclin-1 by using qPCR.Furthermore,adult zebrafish were amputated and randomly divided into the control group,the 3-MA injected group and the STF-62247 injected group.The autophagy inhibitor 3-MA or the activator STF-62247 was injected for 7 consecutive days.The length of regenerated zebrafish caudal fin was recorded every day.The effects of autophagy inhibition or autophagy activation on the regeneration rate of zebrafish caudal fin were analyzed.Results:During the caudal fin regeneration,the expression of ATG and Beclin-1 was down-regulated by 3-MA,while STF-62247 enhanced ATG and Beclin-1 expression levels.Compared with the control group,both inhibition of autophagy and activation of autophagy showed no significant effects on the rate of zebrafish caudal fin regeneration.Conclusion:Through intraperitoneal injection,neither autophagy inhibitor 3-MA nor autophagy activator STF-62247 exhibits any significant effects on the regeneration rate of zebrafish caudal fin.
