摘要
目的 探究白三烯受体拮抗剂对过敏性鼻炎模型大鼠血清白细胞介素-6(Interleukin-6,IL-6)、白细胞介素-10(Interleukin-10,IL-10)及肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)表达的影响。方法 选择SD健康雄性大鼠30只,其中10只不作处理,作为本次研究正常大鼠组,另外20只建立过敏性鼻炎模型,将建模成功的大鼠分为模型大鼠组和白三烯受体拮抗剂组。白三烯受体拮抗剂组每日注射孟鲁司特溶液,正常大鼠组和模型大鼠组大鼠注射等体积0.9%氯化钠溶液。观察HE染色的病理情况,采用动物肺功能仪检测大鼠肺功能:25%最大呼气流量(FEF25)、50%最大呼气流量(FEF50)、75%最大呼气流量(FEF75)、最大呼气中期流量(MMF)、用力最大呼气流量(PEF),酶联免疫吸附测定法检测IL-6、IL-10、TNF-α表达水平。Western blot检测细胞间黏附分子-1(intercellular adhesion molecule-1,sICAM-1)、白细胞三烯D4(leukotriene D4,LTD4)、5-过氧羟基-花生四烯酸(5-hydroperoxyeicosatetraenoicacid,5-HPETE)水平。结果 建模成功16只,模型大鼠组行为学特征总分显著高于正常大鼠组和白三烯受体拮抗剂组,差异有统计学意义(P<0.05)。模型大鼠组FEF25、FEF50、FEF75、MMF、PEF显著低于正常大鼠组(P<0.05)。白三烯受体拮抗剂组FEF25、FEF50、FEF75、MMF、PEF高于模型大鼠组;模型大鼠组血清IL-6、IL-10、TNF-α水平均显著高于正常大鼠组和白三烯受体拮抗剂组大鼠。模型大鼠组sICAM-1、LTD4、5-HPETE水平高于正常大鼠组和白三烯受体拮抗剂组,差异有统计学意义(P<0.05)。结论 白三烯受体拮抗剂应用于过敏性鼻炎模型大鼠,能够有效调控大鼠血清炎症因子IL-6、IL-10及TNF-α的表达,具有抑制炎症细胞增殖,诱导炎症细胞凋亡的作用。
Objective To investigate the effects of leukotriene receptor antagonists on the expression levels of serum interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α)in the rat models with allergic rhinitis.Methods Thirty healthy male SD rats were enrolled in the study,in wgich,10 rats were not treated as control group,the other 20 rats were used to establish the animal models with allergic rhinitis,then the successfully modeled rats were divided into model group and leukotriene receptor antagonist group.The rats in leukotriene receptor antagonist group were injected daily with montelukast solution,and rats in control group aand model group were injected with equal-volume physiological saline.The pathological conditions of HE staining were observed,and the lung function of rats was detected by animal lung function instrument including 25%maximum expiratory flow(FEF25),50%maximum expiratory flow(FEF50),75%maximum expiratory flow(FEF75),MMF,maximum forced expiratory flow(PEF),moreover the expression levels of and IL-6,IL-10,TNF-αwere detected by enzyme-linked immunosorbent assay(ELISA).In addition the expression levels of intercellular adhesion molecule-1(sICAM-1),leukotriene D4(LTD4),5-hydroperoxyeicosatetraenoicacid(5-HPETE)were detected by Western Blot.Results The animal models with allergic rhinitis were successfully established in 16 rats.The total scores of behavioral characteristics in mode group were significantly higher than those in control group and leukotriene receptor antagonist group(P<0.05).The levels of FEF25,FEF50,FEF75,MMF and PEF in model group were significantly lower than those in control group(P<0.05),which in leukotriene receptor antagonist group were higher than those in model group.The serum levels of IL-6,IL-10,TNF-αin model group were significantly higher than those in control group and leukotriene receptor antagonist group.Moreover the levels of sICAM-1,LTD4,5-HPETE in model group were significantly higher than those in control group and leukotriene receptor antagonist group(P<0.05).Conclusion The application of leukotriene receptor antagonist in rat models with allergic rhinitis can effectively regulate the expressions of serum inflammatory factors including IL-6,IL-10 and TNF-α,which can inhibit the proliferation of inflammatory cells and induce apoptosis of inflammatory cells.
