摘要
目的研究非小细胞肺癌(NSCLC)癌组织中微小RNA(miR)-214及MIR31HG的表达及其临床意义。方法回顾性分析2016年2月-2017年2月于中国医科大学附属盛京医院胸外科行手术治疗的NSCLC患者104例,应用荧光实时定量PCR检测NSCLC癌组织和癌旁组织中miR-214与MIR31HG的表达,分析组间miR-214与MIR31HG的表达差异及其与临床病理特征之间的关系,采用ROC曲线分析miR-214与MIR31HG对NSCLC的预测价值。结果 NSCLC癌组织与癌旁组织的miR-214相对表达量分别为(0.37±0.08)、(1.15±0.09),MIR31HG的相对表达量分别为(8.15±1.15)、(1.33±0.31)两者具有统计学意义(t/P=64.877/0.000,58.230/0.000)。癌组织中miR-214及MIR31HG表达与肿瘤分期、组织学分级有关(tmiR-214=12.462、13.834,P=0.000、0.000;tMIR31HG=11.573、12.278,P=0.000、0.000),而与患者病理类型、肿瘤直径及是否存在淋巴结转移无关(P>0.05)。癌组织中miR-214与MIR31HG表达呈负相关(r=-0.618,P=0.000)。miR-214预测NSCLC的敏感度为0.823,特异度为0.772;MIR31HG预测NSCLC的敏感度为0.807,特异度为0.854。结论 NSCLC癌组织中miR-214表达降低,MIR31HG表达升高,二者可能与NSCLC的发生发展有关,有希望成为预测NSCLC发生、发展的肿瘤标志物。
Objective To study the expression of microRNA-214 and MIR31 HG in non-small cell lung cancer(NSCLC)and its clinical significance.Methods A retrospective analysis was made of 104 patients with NSCLC who underwent thoracic surgery from February 2016 to February 2017 in Shengjing Hospital Affiliated to China Medical University.The expression of microRNA-4 and MIR31 HG in NSCLC cancer tissues and adjacent tissues was detected by real-time fluorescence quantitative PCR.The difference of microRNA-4 and MIR31 HG expression between groups and its relationship with clinicopathological characteristics were analyzed.The ROC curve was used to analyze the predictive value of miR-214 and MIR31 HG to NSCLC.Results The relative expression levels of miR-214 in NSCLC and adjacent tissues were(0.37 ±0.08)and(1.15 ±0.09),respectively.The relative expressions of MIR31 HG were(8.15 ± 1.15)and(1.33 ±0.31),respectively.Compared with NSCLC,the expression of miR-214 was lower,while the expression of MIR31 HG was higher(t/P =64.877/0.000,t/P =58.230/0.000).The expression of miR-214 and MIR31 HG in cancer tissues was correlated with tumor stage and histological grade(miR-214:t= 12.462,13.834, P= 0.000,0.000;MIR31 HG:t= 11.573,12.278,P= 0.000,0.000),and the pathological type and tumor diameter of patients There was no correlation with lymph node metastasis(P>0.05).There was a negative correlation between miR-214 and MIR31 HG expression in cancer tissues(r=-0.618,P=0.000).The sensitivity of miR-214 for predicting NSCLC was 0.823 and the specificity was 0.772.The sensitivity of MIR31 HG for predicting NSCLC was 0.807 and the specificity was 0.854.Conclusion The expression of microRNA-214 decreased and MIR31 HG increased in NSCLC tissues.Both of them may be related to the occurrence and development of NSCLC,and may be a tumor marker for predicting the occurrence and development of NSCLC.
