摘要
目的研究决奈达隆和胺碘酮对肝脏的影响及多烯磷脂酰胆碱的保护作用。方法将SD雄性大鼠50只分为胺碘酮组(AM组)、决奈达隆组(DR组)、胺碘酮+多烯磷脂酰胆碱组(AC组)、决奈达隆+多烯磷脂酰胆碱组(DC组)及对照组。胺碘酮和决奈达隆药物剂量均为120mg·kg^-1·d^-1,多烯磷脂酰胆碱剂量为200mg·kg^-1·d^-1,给药13周。分别于第4、8、12、13周末取血,检测丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(AKP)、总蛋白(TP);处死大鼠后取肝脏行病理组织检查。结果 AM组和DR组ALT、AST水平于第8周[AM组:ALT(60.46±7.73)U/L、AST(134.46±20.49)U/L,DR组:ALT(65.67±7.52)U/L、AST(137.16±21.19)U/L]和第12周[AM组:ALT(50.08±8.88)U/L、AST(98.81±9.62)U/L,DR组:ALT(63.17±14.11)U/L、AST(105.13±16.20)U/L]升高,与对照组[ALT(55.95±12.25)U/L、AST(114.15±14.90)U/L;ALT 43.32±10.02U/L、AST 79.86±16.06U/L]比较差异均有统计学意义(均P<0.05),AC、DC组分别与AM组、DR组比较ALT及AST水平呈下降趋势(P<0.05)。电镜显示AM组及DR组肝脏细胞空泡及线粒体结构改变。结论多烯磷脂酰胆碱能拮抗胺碘酮和决奈达隆科引起的肝功能异常。
Objective To explore the influence of dronedarone and amiodarone on hepatic function and the protective effect of polyene phosphatidylcholine. Methods 50 male SD rats were randomly divided into 5 groups(10 in each group):normal control(NC), amiodarone(AM), dronedarone(DR), amiodarone with polyene phosphatidylcholine(AC), and dronedarone with polyene phosphatidylcholine(DC) groups. The dosage of both amiodarone and dronedarone was 120 mg/kg/d. The dosage of polyene phosphatidylcholine was 200 mg/kg/d. The blood was sampled for measuring alanine aminotransferase(ALT), aspartate aminotransferase(AST), alkaline phosphatase(AKP) and total protein(TP) at the end of 4, 8, 12, and 13 weeks. All rats were sacrificed at the end of 13 weeks and the hepatic organs were isolated for histopathological examination. Results ALT and AST levels in AM group(60.46 ±7.73 U/L and 134.46 ±20.49 U/L at 8 th week, 50.08±8.88 U/L and 98.81±9.62 U/L at 12 nd week) and DR group(65.67 ±7.52 U/L and 137.16 ±21.19 U/L at 8 th week, 63.17 ±14.11 U/L and 105.13 ±16.20 U/L 12 nd week) were significantly higher than those in NC group(55.95 ±12.25 U/L and 114.15±14.90 U/L at 8 th week, 43.32 ±10.02 U/L and 79.86 ±16.06 U/L 12 nd week)(all P<0.05). ALT and AST levels were significantly lower in AC and DC groups than in AM and DR groups(P < 0. 05). Electron micrograph showed hepatic cell foam and structural changes of mitochondria in AM and DR groups. Conclusion Polyene phosphatidylcholine could protect rat against amiodarone-and dronedarone-induced hepatic injury.
