摘要
目的:筛选对多重耐药大肠杆菌抑制作用较好的中药,进行组方,研发新药,探讨中药提取物或中药制剂对防治多重耐药菌株引起疾病的效果。方法:采用纸片扩散法(K-B法)检测不同来源的大肠杆菌对13种抗生素类药物的敏感性,筛选出多重耐药菌株;采用牛津杯法测定45味中药提取液对多重耐药菌的抑菌圈直径,筛选出表现为高敏的中药;采用二倍稀释法确定10味高敏中药的最小抑菌浓度,评估不同中药对多重耐药菌株的抑制效果。结果:筛选出耐药率高于70%的多重耐药菌株7株,其中猪源3株、羊源1株、鹅源3株;多重耐药菌株对28味中药表现为不敏感,对7味中药表现为中敏,对10味中药表现为高敏;抑菌效果最好的3味中药依次是博落回、黄连和水杨梅。结论:不同中药对同一多重耐药菌株抑制效果不同,一些中药对多重耐药菌可以起到较好的体外抑制效果,为研制临床上耐药性疾病防治的新药物提供参考。
Objective: To screen some Chinese medicines with strong inhibitory effect on multidrug-resistant E.coli and formulate the prescription,develop new drugs,and discuss the effect of using Chinese herbal medicine extract or Chinese herbal medicine preparation to prevent and cure the disease caused by multi-drug-resistant strains. Methods: K-B method was applied to test the susceptibility of E.coli to 13 kinds of antibiotics and screen out the multi-drug resistant strains.Oxford cup method was used to determine the inhibition zone diameter of 45 Chinese herbal extracts against multidrug-resistant bacteria and screen out highly sensitive Chinese herbs.Two fold dilution method was used to determine the minimal inhibitory concentration (MIC )of the 10 highly sensitive Chinese herbs and evaluate the inhibitory effect of different Chinese medicines on multidrug- resistant strains. Results: A total of 7 strains of multidrug-resistant strains with a resistance rate of more than 70% were screened out.Among them,3 strains were from pigs,1 from sheep and 3 from goose.Multidrug-resistant strains showed resistance to 28 Chinese medicines,medium sensitivity to 7 Chinese herbs and highly sensitive to 10 herbs.The best 3 antibacterial herbs were Macleaya cordata , Coptis chinensis and Myrica rubra . Conclusion: Different Chinese medicines have different inhibitory effects on the same multidrug-resistant bacteria.Some Chinese medicines exhibit better inhibitory effects on multidrug-resistant bacteria in vitro ,which provides a basis for the development of new drugs for the prevention and treatment of clinical drug-resistant diseases.
