摘要
目的:探究长链非编码RNA脑细胞质RNA1(BCYRN1)在结直肠癌组织中的表达特征及其与细胞侵袭转移的关系。方法:35例结直肠癌组织及相应的癌旁正常组织收集于2016年3月至2017年12月期间经我院普外科行结直肠癌根治术的患者,采用实时荧光定量PCR(Real-time fluorescence quantitative PCR,RT-qPCR)技术检测结直肠癌及癌旁正常的组织中BCYRN1、Snail及E-cadherin的mRNA表达水平;比较不同来源组织中BCYRN1、Snail及E-cadherin表达差异并分析BCYRN1与Snail及E-cadherin表达的相关性;分析BCYRN1表达与患者病理资料间的关系。采用过表达载体慢病毒转染技术外源性上调结直肠癌细胞SW480中BCYRN1的表达以此作为实验组细胞,转染空白对照载体的SW480作为对照组细胞,Transwell侵袭及迁移实验检测细胞转移能力;RT-qPCR技术检测Snail及E-cadherin mRNA表达的变化;Western Blot实验检测Snail及E-cadherin蛋白表达水平的变化。结果:实验组Transwell基底膜侵袭及迁移细胞数显著高于对照组,实验组BCYRN1及Snail mRNA表达显著高于对照组,E-cadherin mRNA表达显著低于对照组,Snail蛋白表达显著高于对照组,E-cadherin蛋白表达显著低于对照组。BCYRN1及Snail mRNA在结直肠癌组织中表达显著高于癌旁正常的组织,E-cadherin mRNA在结直肠癌组织中表达显著低于癌旁正常的组织,结直肠癌组织中BCYRN1与Snail mRNA表达显著正相关,与E-cadherin mRNA表达显著负相关,结直肠癌组织中BCYRN1高表达患者TNM分期及浸润深度显著高于低表达患者。结论:BCYRN1可通过Snail/E-cadherin途径促进结直肠癌侵袭转移能力。
Objective:To investigate the expression of long non-coding RNA BCYRN1 in colorectal cancer and its relationship with cell invasion and metastasis.Methods:35 cases of colorectal cancer tissues and corresponding normal para-cancerous tissues of 35 cases were collected in the Department of General Surgery of our hospital during 2016.3-2017.12.The expression of BCYRN1,Snail and E-cadherin mRNA in colorectal cancer and normal para-cancer tissues were detected by Real-time fluorescence quantitative PCR(RT-qPCR).The expression of BCYRN1,Snail and E-cadherin in different tissues were compared and their correlations were analyzed.The relationship between BCYRN1 expression and pathological data were analyzed.The expression of BCYRN1 of colorectal cancer cells SW480 which was up-regulated by overexpression vector lentivirus transfection was as test group,and SW480 transfected blank control vector was as control group.Cell metastasis ability was detected by Transwell invasion and migration test.The changes of Snail and E-cadherin mRNA expression were detected by RT-qPCR technique.The changes of Snail and E-cadherin protein expression level were detected by Western Blot.Results:The number of cell that invaded and migrated in Transwell basement membrane of the test group was significantly higher than that of the control group.The expression of BCYRN1 and Snail mRNA in the test group was significantly higher than that in the control group.The expression of E-cadherin mRNA was significantly lower than that of the control group.The expression of Snail protein was significantly higher than that of the control group,and the expression of E-cadherin protein was significantly lower than that of the control group.The expression of BCYRN1 and Snail mRNA in colorectal carcinoma was significantly higher than that of normal para-cancerous tissue.The expression of E-cadherin mRNA in colorectal carcinoma was significantly lower than that of normal para-cancerous tissue.The expression of BCYRN1 in colorectal cancer tissues was positively correlated with the expression of Snail mRNA,and was negatively correlated with the E-cadherin mRNA.TNM stage and infiltration depth in patients with high expression of BCYRN1 in colorectal cancer were significantly higher than those in patients with low expression of BCYRN1.Conclusion:BCYRN1 can promote the invasion and metastasis of colorectal cancer through Snail/E-cadherin pathway.
