响应面法优化裂殖壶菌产DHA油脂的发酵培养基

Medium optimization of Schizochytrium sp. for DHA oil production by response surface methodology

摘要 采用响应面法对海洋真菌Schizochytrium sp.产DHA油脂的发酵培养基进行优化。通过单因素试验确定了以葡萄糖为碳源、酵母浸粉为氮源、谷氨酸钠为外源添加剂3个主要因素。根据Box-Behnken中心组合设计,以DHA油脂产量为响应值,对Schizochytrium sp.DP-16的发酵培养基进行响应面优化。优化后的培养基组成为葡萄糖83.60 g/L,酵母浸粉11.75 g/L,谷氨酸钠9.30 g/L,海水晶15 g/L,MgSO4·7H2O 5 g/L,KH2PO4·H2O 7 g/L。优化后DHA油脂产量提高了4.5%。 The fermentation medium for DHA oil production by marine fungus Schizochytrium sp.was optimized by response surface methodology(RSM).Three main factors were determined through single factor test:glucose as carbon source,yeast extract as nitrogen source and sodium glutamate as exogenous additive.According to Box-Behnken central combination design,the fermentation medium for DHA production was optimized by RSM.The optimum medium composition were as follows(g/L):glucose 83.60,yeast extract 11.75,monosodium glutamate 9.30,sea salt 15,MgSO4·7H2O5,KH2PO4·H2O7.DHA oil yield was increased by 4.5%after optimized.
