摘要
优化复合酶解法辅助提取香菇菌糠多糖的提取工艺,并研究其抗氧化活性。通过测定清除DPPH自由基、羟自由基(·OH)和超氧阴离子(O2-·)的能力评价香菇菌糠多糖的抗氧化活性。结果表明,复合酶解法提取香菇菌糠多糖最佳提取条件为:酶解时间50 min,酶解温度45℃,pH值5.0,酶浓度3%,在此提取条件下香菇菌糠多糖的提取率为9.60%,显著优于传统的热水浸提法,提取率提高了52.70%。通过复合酶解法提取的香菇菌糠多糖清除羟自由基的半数抑制浓度(IC50)为0.58 mg/ml、清除超氧阴离子的IC50为0.60 mg/ml,清除DPPH自由基的IC50为0.90 mg/ml。表现出较强的抗氧化活性,且抗氧化活性随多糖浓度的增加而升高。
The extraction process of bran polysaccharide fromLentinus edodes residue by complex enzymatic hydrolysis was optimized and its antioxidant activity was studied. The antioxidative activity of polysaccharide of Lentinus edodes residue was evaluated by measuring the ability of removing DPPH free radical, hydroxyl free radical(·OH) and superoxide anion (O2^-·) The results showed that the optimal extraction conditions of polysaccharide by the compound enzyme method were: enzymatic hydrolysis time 50 min, enzymolysis temperature 45 ℃, pH 5.0, enzyme concentration 3%. Under the conditions, the extraction yield of polysaccharide was 9.60%, significantly better than the traditional hot water extraction, extraction yield increased by 52.70%. The IC50 of Lentinus edodes residue polysaccharides extracted by complex enzymatic hydrolysis was 0.58 mg/ml for scavenging hydroxyl radicals, 0.60 mg/ml for scavenging superoxide anions,and 0.90 mg/ml for scavenging DPPH radicals. It showed strong antioxidant activity, and the antioxidant activity increased with the increase of polysaccharide concentration.
