摘要
目的建立HPLC同时测定蒙药阿那日-4中鞣花酸、桂皮醛和胡椒碱3种活性成分含量的方法。方法采用Agilent ZORBAX Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 ml/min,柱温30℃,检测波长271 nm。结果供试品溶液在24 h内稳定,鞣花酸、桂皮醛、胡椒碱质量浓度分别在0.050~0.500,0.030~0.300,0.030~0.300 mg/ml范围内与峰面积呈良好的线性关系,相关系数分别为0.9994,0.9995,0.9997;平均加样回收率(n=6)分别为101.68%,100.94%,98.55%,RSD分别为1.806%,1.415%,1.790%。结论该方法简便准确,专属性强,重复性好,可用于蒙药阿那日-4的质量控制方法。
Objective To establish an HPLC method for the simultaneous determination of ellagic acid, cinnamaldehyde and piperine in Anari-4. Methods The chromatographic separation was carried out on an Agilent ZORBAX Eclipse XDB-C18 column (250 mm×4.6 mm, 5 μm) with the mobile phase consisting of methanol-0.1 %(v/ v) phosphoric acid solution in a gradient mode at the flow rate of 1.0 ml/min. The detection wavelength was set at 271 nm and the column temperature was 30 ℃. Results The sample solution was stable within 24 h, the method showed good linear relationship within the range of 0.050-0.500 mg/ml for ellagic acid (r=0.9994), 0.030-0.300 mg/ml for cinnamaldehyde (r=0.9995) and 0.030-0.300 mg/ml for piperine (r=0.9997);the average recoveries were 101.68 %, 100.94 % and 98.55 %, respectively;and the RSDs were 1.806 %, 1.415 % and 1.790 %, respectively. Conclusion The method is handy and accurate, with strong specificity, and could be used as the quality control of mongolian medicine Anari-4.
