摘要
目的:探讨白藜芦醇(Res)的抑瘤作用及其机制,为其开发应用提供实验依据。方法:通过右腋窝皮下接种Lewis肺癌细胞悬液的方法制备荷瘤小鼠模型30只,将其随机分为模型组、Res组和环磷酰胺组,各10只,另取10只小鼠为正常对照组。正常对照组、模型组均给予生理盐水灌胃,环磷酰胺组腹腔注射环磷酰胺,Res组给予Res灌胃给药。比较各组生长情况、瘤重、脾淋巴细胞转化率(SI值)、血清IL-2和IL-4水平。结果:与模型对照组比较,Res组小鼠皮毛松散、饮食差等状况减轻;Res组瘤重低于模型组(P<0.05);模型组和环磷酰胺组SI值均低于正常对照组、Res组(P<0.05);模型组、环磷酰胺组IL-2水平均低于正常对照组、Res组,IL-4水平均高于正常对照组、Res组,比较差异均有统计学意义(P<0.05);但Res组与正常对照组SI值、IL-2、IL-4水平比较,差异均无统计学意义(P>0.05)。结论:Res能够抑制小鼠体内Lewis肺癌细胞生长,其机制可能与调节体内细胞因子水平,纠正荷瘤机体的Th1/Th2漂移现象有关。
Objective:To explore the anti-tumor effect and mechanism of resveratrol(Res)and provide experimental basis for its development and application.Method:A total of 30 mice bearing Lewis lung cancer were prepared by subcutaneous inoculation of Lewis lung cancer cell suspension in the right axilla,they were randomly divided into model group,Res group and cyclophosphamide group,10 rats in each group,another 10 mice were taken as normal control group.Normal control group and model group were given saline intragastric administration,Cyclophosphamide group was given Cyclophosphamide intraperitoneally,Res group was given Res intragastric administration.The growth,tumor weight,splenic lymphocyte transformation rate(SI value),serum IL-2 and IL-4 levels in each group were compared.Result:Compared with the model group,the mice in Res group had less loose fur,poor diet.The tumor weight of Res group was lower than that of model group(P<0.05).The SI values of model group and Cyclophosphamide group were lower than those of normal control group and Res group(P<0.05).The level of IL-2 in model group and Cyclophosphamide group were lower than those of normal control group and Res group,and the level of IL-4 were higher than those of normal control group and Res group,the differences were significant(P<0.05).However,the SI,IL-2 and IL-4 levels in Res group and normal control group were compared,the differences were not significant(P>0.05).Conclusion:Res can inhibit the growth of Lewis lung cancer cells in mice,and its mechanism may be related to regulating the level of cytokines in vivo and correcting the Th1/Th2 drift phenomenon in tumor-bearing organisms.
