摘要
目的:观察定喘汤对RSV感染大鼠TSLP蛋白表达及IL-4、IL-10、IL-13含量的变化,探讨定喘汤对RSV感染大鼠后Th2应答反应上、下游因子的影响机制。方法:将Wistar大鼠随机分为正常组、RSV模型组和定喘汤全方组,每组各9只,除正常组外,其余各组用RSV(Long珠)病毒滴鼻感染大鼠,感染后正常组和RSV模型组均以等体积的生理盐水灌胃,定喘汤组予同等量的中药灌胃。在感染后7d,取大鼠肺组织行病理分析,采用蛋白印迹法(WB)和SABC免疫组化方法检测大鼠肺组织TSLP蛋白表达的情况,同时应用酶联免疫吸附试验(ELISA)检测大鼠血清中Th2型相关细胞因子IL-4、IL-10和IL-13的表达水平。结果:与正常组比较,模型组大鼠肺组织中TSLP蛋白表达及血清中的IL-4、IL-10、IL-13含量增高具有显著性差异(P<0.05);与模型组比较,定喘汤组TSLP蛋白表达及血清中的IL-4、IL-10、IL-13含量显著减弱(P<0.05)。结论:定喘汤干预RSV感染大鼠的作用机制可能与抑制大鼠Th2上游因子TSLP蛋白表达及下游因子IL-4、IL-10、IL-13有关。
Objective:To observe the expression of TSLP protein and the levels of IL-4,IL-10 and IL-13 in rats with RSV infection and explore the mechanism of Dingchuan Decoction on the up and downstream factors of Th2 response in RSV infected rats.Methods:We divided Wistar rats equally into 3 groups randomly:normal group,RSV model group,Dingchuan Decoction group,9 rats in each group.Except the normal group,the rats were infected with RSV(Long bead)virus and the rats in the normal group and the RSV model group were given equal volume of normal saline,and the Dingchuan Decoction group was treated with the corresponding drugs by gavage.Seven days after infection,the lung tissue of rats was examined by pathology.The levels of TSLP protein in lung tissue were detected by Western blotting and immune histochemistry.The levels of IL-4,IL-10 and IL-13 in Th2-related cytokines were measured by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal group,the expression of TSLP protein in lung tissue and IL-4,IL-10 and IL-13 in serum were significantly higher in the RSV model group(P<0.05).The expression of TSLP protein and IL-4,IL-10,IL-13 were down-regulated in the Dingchuan Decoction group compared with those of RSV model group(P<0.05).Conclusion:The mechanism of Dingchuan Decoction on RSV infection in rats may be related to the inhibition of Th2 upstream TSLP protein expression and downstream factors IL-4,IL-10,IL-13.
