摘要
为了研究创建薄皮甜瓜‘花雷’的再生体系,以薄皮甜瓜‘花雷’为试材,子叶、胚轴、真叶为外植体,观察其在含有不同种类和浓度生长调节剂的培养基中的再生频率。诱导愈伤组织的最佳外植体为子叶,最佳诱导培养基为MS+0.5 mg·L^(-1)6-BA+0.5 mg·L^(-1)IAA,最佳分化培养基为MS+0.5 mg·L^(-1)6-BA+0.2 mg·L^(-1)IBA,最佳生根培养基为MS+0.2 mg·L^(-1)IBA。初步选出一套完整的适宜薄皮甜瓜‘花雷’再生体系建立的培养基。
The regeneration system of‘Hualei’of melon was established.The regeneration frequency in culture medium containing different kinds and concentration of growth regulator was observed by using‘Hualei’of melon as the experimental material and cotyledon,hypocotyl and true leaf as explants.The best explants for callus induction were cotyledon,the best induction medium was MS+0.5mg·L^-16-BA+0.5mg·L^-1IAA,the best differentiation medium was MS+0.5 mg·L-1 6-BA+0.2mg·L-1 IBA,and the best rooting medium was MS+0.2 mg·L-1 IBA.A complete culture medium suitable for the regeneration system of the melon‘Hualei’was selected.
