环境内分泌干扰物对金鱼性腺组织学影响 被引量:6

The effect of environmental endocrine disruptors on the gonad histology of Carassius auratus

摘要 [目的]"环境内分泌干扰物"又称"环境激素",近年来由于其对环境造成的污染逐渐增加,受到国内外广泛关注。本文选取市场上常见的金鱼作为试验材料,研究3种环境激素对金鱼性腺组织学的影响。[方法]甲基睾酮(17α-methyltestosterone, MT)是一类人工合成的雄激素,乙炔雌二醇(Ethinylestradiol, EE2)是雌激素干扰物中的一个典型代表,来曲唑(Letorzole, LE)是新一代芳香化酶抑制剂,本文分别设置暴露3、7 d的0、50、100、200 ng·L^(-1)的MT浓度梯度;暴露3、7 d的0、50、100、200 ng·L^(-1)的EE2浓度梯度;暴露7、14 d的0、100、200、400 ng·L^(-1)的LE浓度梯度。[结果]发现MT对金鱼卵巢具有抑制作用;不同EE2浓度处理的雄性金鱼体重和性腺重均受到了抑制,50 ng·L^(-1)的EE2处理金鱼7 d后,变化最为明显,不同EE2浓度处理的雌性金鱼体重和性腺重均受到了促进,100 ng·L^(-1)的EE2处理雌性金鱼7 d后,变化最为明显; LE对于雌鱼的卵巢分化具有抑制作用,并且在100 ng·L^(-1)处理7 d后,抑制作用最为明显,一定浓度的来曲唑可以促进精巢发育,在中等浓度200 ng·L^(-1)处理金鱼7 d时促进作用最为明显。[结论]用3种浓度的EDCs处理金鱼后,MT可抑制金鱼卵巢的发育;EE2可抑制雄性金鱼睾丸的发育,促进雌性金鱼卵巢的发育;LE可抑制雌鱼卵巢的分化,促进睾丸的发育。以上结果显示,3种EDCs可对雄性或雌性金鱼性腺的发育产生不同的干扰作用,且与所用的剂量和暴露时间有关。 [Objective]In recent years,environmental endocrine disruptors,also called"environmental hormones",have attracted wide attention from both domestic and globe researches due to their negative impact on environmental pollution development.As a typical,common and representative model,goldfish Carassius auratus was selected as experimental material to explore the effect of three environmental hormones on the gonad histology.[Methods]17α-methyltestosterone(MT)is a synthetic steroid hormone.Ethinylestradiol(EE2)is a typical representative of estrogen disruptors,and Letorzole(LE)is a new generation of aromatase inhibitors.These three chemicals were employed as endocrine disrupting chemicals(EDCs)in the present study.Three concentrations of both MT and EE2,50,100 or 200 ng·L-1,were applied,and exposure time was set up for 3 and 7 days.A gradient concentrations of LE were made as 0,100,200,and 400 ng·L-1,and the effect of LE was measured at 7 and 14 days post treatment.[Results]Results showed that MT inhibited goldfish ovary development 7 days after exposure.During 7 days exposure time,both body weight and gonad weight of male goldfish were suppressed when treated with EE2 at different concentrations with the highest effect observed at 50 ng·L-1 treatment.In contrast,both body weight and gonad weight of female goldfish were promoted at different concentration of EE2,especially at 100 ng·L-1 after 7 days post treatment.The inhibitory activity of LE on differentiation of female ovarywas observed,especially at 100 ng·L-1 LE treatment at 7 and 14 days post treatment,while testis development was promoted at certain concentrations,especially at 200 ng·L-1 after 7 days exposure time.[Conclusion]In summary,when goldfish was treated with three concentrations of EDCs,MT inhibited the development of goldfish ovary,and EE2 suppressed the testis development of male goldfish and promoted the development of female goldfish ovary.LE slowed down the differentiation of female ovary,while promoted testis development.The obtained results indicated that three EDCs can play different interference effects on the development of male or female goldfish gonads,and relate to the dose and time used.
