幽门螺杆菌对人组织淋巴瘤细胞的损伤作用及机制 被引量:1

摘要 目的观察幽门螺杆菌(Hp)对人组织淋巴瘤细胞的损伤作用,探讨Hp诱导的胃黏膜慢性炎症反应机制。方法取人组织细胞淋巴瘤细胞系U937,分为Hp组和对照组。Hp组按细菌与细胞100∶1的比例加入Hp,对照组加入等体积RPMI1640培养液,培养14 h。采用ELISA法检测两组细胞培养上清中炎症因子巨噬细胞移动抑制因子(MIF)、白细胞介素1β(IL-1β)水平; RT-PCR法检测细胞中MIF、IL-1β、核因子κB抑制蛋白α(IκB-α)、核因子κB(NF-κB) mRNA的相对表达量; CCK-8法检测细胞增殖情况,判断细胞毒性; Western boltting法检测细胞内IκB-α、NF-κB p65蛋白表达。结果 Hp组细胞培养上清中MIF、IL-1β水平较对照组升高(P均<0. 05),细胞中MIF、IL-1β、IκB-α、NF-κB mRNA表达较对照组升高(P均<0. 05),细胞增殖的OD值降低(P <0. 01),IκB-α、NF-κB p65蛋白表达较对照组升高(P均<0. 05)。结论 Hp感染人组织淋巴瘤细胞后可诱导相关炎症因子分泌增加,激活NF-κB信号通路,进而促进人组织淋巴瘤细胞的炎症损伤,扰乱机体免疫防御反应,导致胃黏膜慢性炎症的发生发展。 Objective To investigate the damage effect of Helicobacter pylori(Hp)on human histiocytic lymphoma cells and the mechanism of cell-associated inflammatory reaction induced by Hp and its damage to host U937 cells.Methods The human histiocytic lymphoma cell line U937 was divided into the Hp group and control group.The levels of macrophage migration inhibitory factor(MIF)and interleukin-1β(IL-1β)in the culture supernatants of the above two groups were detected by ELISA.The relative expression levels of MIF,IL-1β,IκB-αand NF-κB mRNA in the above two groups were detected by RT-PCR.The cytotoxicity of cells in the two groups was detected by CCK-8;the expression levels of IκB-αand NF-κB p65 protein in the above two groups were analyzed by Western blotting.Results The levels of MIF and IL-1βin the cell culture supernatant of the Hp group were higher than those in the control group(both P<0.05).The expression levels of MIF,IL-1β,IκB-αand NF-κB mRNA in the HP group were higher than those in the control group(all P<0.05),the cell proliferation ability decreased(P<0.01),and the expression of IκB-αand NF-κB p65 protein in the HP group was higher than that of the control group(both P<0.05).Conclusion Hp infection of human histiocytic lymphoma cells can induce the increased secretion of related inflammatory factors,activate NF-κB signaling pathway,and thereby promote inflammatory damage of human tissue lymphoma cells,disrupt the body's immune defense response,and lead to the development of chronic inflammation of gastric mucosa.
