摘要
测定了柔鱼亚科(Ommastrephinae)中的柔鱼(Ommastrephes bartramii)、茎柔鱼(Dosidicus gigas)、鸢乌贼(Sthenoteuthis oualaniensis)、发光柔鱼(Eucleoteuthis luminosa)的线粒体COI和16S rRNA基因的部分序列,结合从Gen Bank中获得的玻璃乌贼(Hyaloteuthis pelagica)、鸟柔鱼(Ornithoteuthis volatilis)、大西洋鸟柔鱼(Ornithoteuthis antillarum)的同源序列,以太平洋褶柔鱼(Todarodes pacificus)、阿根廷滑柔鱼(Illex argentinus)为外群,运用贝叶斯法(BI)构建了分子系统树,同时联合了2个基因片段序列,运用贝叶斯联合模型综合探讨了柔鱼亚科种类的系统发育关系。研究发现,除16S rRNA基因外,COI基因片段和联合模型所构建的系统树拓扑结构完全一致;结合形态学资料,发现玻璃乌贼、发光乌贼和鸢柔鱼亲缘关系较近,且形态上都出现了外套膜大型发光器,是柔鱼亚科中分化较晚的种类;其次茎柔鱼、柔鱼与其它种类存在较大的遗传距离,形态上丢失了内脏和眼球发光器,可能预示着柔鱼亚科种类一个新的进化方向;鸟柔鱼属两个种为柔鱼亚科中最原始种类,且为褶柔鱼亚科向柔鱼亚科进化过程中的过渡物种。
Ommstrephinae represents a relatively small subfamily of Cephalopoda with highly various morphology,which includes 6 genera and 9 species.Many species in the subfamily are well studied due to their ecological importance and commercial value.Several phylogenies of Ommstrephinae based on morphological and molecular data can be found,while the phylogenetic placement of Ommstrephinae species and the identification of their closest relatives remains to be controversical.Our study will add important knowledge for understanding the interrelationships of Ommstrephinae and also provide insights into historical biogeography of Ommstrephinae.We investigated the phylogenetic relationships of Ommastrephinae through Partitioned Bayesian Analyses with concatenated genes sequences using two mitochondrial genes.Fragments of 16S rRNA and COI with a total of 1 071 nucleotides were sequenced in Ommastrephes bartramii,Dosidicus gigas,Sthenoteuthis oualaniensis,Eucleoteuthis luminosa,and homologous sequences of Hyaloteuthis pelagica,Ornithoteuthis volatilis,Ornithoteuthis antillarum were downloaded from genbank database,as well as Todarodes pacificus,Illex argentinus as a outgroup.Due to conflicting phylogenetic signal between 16S and COI gene data,we constructred a phylogenetic tree based on Bayesian analyses for two combined mitochondrial genes.The phylogenetic tree based on Bayesian analyses with concatenated genes sequences was in agreement with that based on COI gene.Molecular phylogenetic analysis of data from 2 genes was in agreement with previous hypotheses.One group was composed of all sampled species of Ommstrephinae based on COI and combined data.Both COI gene and combined data supported the monophyly of Ommstrephinae,while monophyletics a subfamily Ommstrephinae was not supported by 16S analysis data.Ornithoteuthis volatilis formed a clade with Ornithoteuthis antillarum to exclusion of Ommstrephinae accroding to 16S data,however the branches of pyhlogenetic tree showed a low posterior probabilities.The topological relationship showed that Sthenoteuthis oualaniensis,Eucleoteuthis luminosa,Hyaloteuthis pelagica and Dosidicus gigas formed a clade,sister-groups of Ommastrephes bartramii.The genera of Ornithoteuthis were placed in the base of phylogenetic tree.The results showed that:The three species of S.oualaniensis,E.luminosa and H.pelagica which had large mantle photophores,showed a closer affinty to D.gigas and O.bartramii,and were the latest species of Ommastrephinae;however,D.gigas and O.bartramii had lost photophores on intestine and eyeball in morphology,suggesting a new direction of evolution of Ommastrephinae;the resulant topology suggested that the two species of the genus Ornithoteuthis were the most likely basal species within Ommastrephinae,which was likely a transitional species from Todarodinae to Ommastrephinae.The results show that photophores on intestine or eyeball play an important role in the phylogentic evolutional of the species of Ommstrephinae.
