兔后肢失用后兰诺定受体与受磷蛋白mRNA转录水平的变化 被引量:1

The Transcription Level Variation of Ryanodine Receptor and Phospholamban mRNA in Rabbits with Muscle Disuse Atrophy

摘要 探讨参与肌浆网中钙离子释放的兰诺定受体(ryanodine receptor,RyR)、受磷蛋白(phospholamban,PLN)的mRNA转录水平与失用性肌萎缩的关系。将依次编号的30只新西兰大白兔按随机数字法分为6组。分别设置为对照组和实验组5组。实验组采用长腿石膏固定法建立兔失用性肌肉萎缩模型,于2周、4周、6周、8周、10周分别处死兔子5只。对照组于第10周处死兔子5只。取固定侧肌肉组织,用定时定量PCR(real-time quantitative polymerase chain reaction,RT-PCR)法测定兔固定侧肌肉的RyR与PLN的基因转录水平。RyR的mRNA转录水平在第2周升高,自第2周开始转录水平逐渐下降。PLN的mRNA转录水平表现为逐渐下降的趋势。在失用性萎缩发展过程中,PLN和RyR mRNA随着肌肉萎缩程度的不断加深呈现趋势性变化,可望用于失用性肌萎缩程度的推断。 The transcription level of ryanodine receptor(RyR)mRNA and phospholamban(PLN)mRNA of disused muscles in rabbits and to explore the relationship with the fixation time were investigated.Thirty New Zealand rabbits were randomly divided into 6 groups(5 rabbits in each group),including five test groups(2,4,6,8,10 weeks)and one control group.The disease model was established by fixing the right rear knee joint in extension position using plaster cast.The rabbits were respectively sacrificed at 2,4,6,8,10 weeks after operation.The five rabbits of control group were sacrificed at the 10th week.The transcription level of RyR and PLN mRNA in the fixed muscles of rabbits was determined by real-time quantitative polymerase chain reaction(RT-PCR).The transcription level of RyR mRNA increased within 2 weeks and then decreased gradually in the following 2 weeks.The transcription level of PLN mRNA decreased gradually.The transcription level of RyR、PLN mRNA shows a time-related change after fixation of the skeletal muscle.It is a potential promising method to evaluate the degree of muscular atrophy.
