摘要
猪瘟E2亚单位疫苗是一种用杆状病毒载体系统表达的猪瘟病毒E2重组蛋白,与油佐剂按比例混合、乳化制成的新型灭活疫苗,可区分感染和接种疫苗动物,有助于猪场的猪瘟净化。为评价该疫苗免疫效力,本研究用猪瘟E2亚单位疫苗与猪瘟兔化弱毒疫苗(传代细胞源)分别免疫靶动物,二免后2 w用强毒人工感染试验动物,连续测温16 d,并记录试验猪临床症状,结合病理剖检、组织器官病毒含量测定结果,比较不同疫苗保护力。结果显示:对照组猪攻毒后体温高热稽留,表现出典型猪瘟症状,攻毒后11 d内5/5发病死亡,尸检剖检呈典型猪瘟病理解剖学变化;免疫猪全部健活,未表现任何猪瘟症状,组织器官剖检无病变。结果表明,猪瘟E2亚单位疫苗与活疫苗保护效力相当,可为靶动物提供可靠的免疫保护效力。
The E2 subunit vaccine emulsified with oil adjuvant is a genetically engineered vaccine based on baculovirus-expressed E2 glycoprotein of Classical swine fever virus(CSFV).The vaccinated pigs with this E2 subunit vaccine can be differentiated from the field virus infected pigs so that the use of this vaccine may help eradicate classical swine fever on pig farms.In the present study,this E2 subunit vaccine and modified live vaccine were used to immunize pigs and antibody responses were examined in ELISA.Two weeks after the second vaccination,all pigs were challenged with CSFV strain Shimen.All of 15 pigs were daily examined for clinical signs and body temperature.The results showed that non-vaccinated pigs displayed typical CSF clinical signs,deaths(5/5)and pathological lesions.All vaccinated pigs survived and did not show obvious signs of CSF.No remarkable pathological changes were observed in vaccinated pigs.These results indicated that the E2 subunit vaccine induced immunoprotection in vaccinated pigs as comparable to the modified live vaccine.
