毛竹根际2株溶磷解钾促生细菌的筛选鉴定 被引量:11

Isolation and Identification of Moso Bamboo Root-Promoting Rhizobacteria and Their Growth-Promoting Effect

摘要 为获得毛竹根际促生细菌(PGPR),评价其对毛竹苗的促生作用。采用稀释平板法从毛竹根际土壤筛选出优良PGPR菌株,测定其溶磷、解钾及分泌吲哚乙酸(IAA)的能力,并对优良菌株进行鉴定;采用回接及盆栽试验研究其对毛竹的促生作用。结果分离到2株优良PGPR菌株JGSB02和JGSB06,其溶磷量分别为165.42,160.29 mg/L;解钾量分别为3.83,3.13 mg/L;产IAA量分别为4.38,10.27 mg/L。形态学特征、生理生化特征和16S r DNA序列分析结果表明,JGSB02和JGSB06菌株均属于Burkholderia cepacia;温室盆栽试验显示JGSB02和JGSB06菌株对毛竹实生苗有显著的促生长作用。进一步研究发现JGSB02和JGSB06菌株对毛竹枯梢病病原菌具有较好的拮抗作用,抑菌率分别为56.67%和66.99%。表明该类细菌可作为促进毛竹生长和生防的潜在PGPR菌株。 In order to obtain PGPR strains,and evaluate their plant growth promoting effect on moso bamboo,phosphate/potassium-solubilizing and IAA-secreting strains were isolated from the rhizosphere soil of moso bamboo,and their growth promoting function on moso bamboo was investigated.The strains were separated from the rhizosphere soil by the method of dilution plate and plate cultivation,and their phosphorus solubilizing ability,k-releasing ability and the capacity of IAA were also determined.The growth promoting effect of the excellent strains on moso bambo was studied using tieback technology and soil pot experiment.The results showed that 2 bacteria were isolated from rhizosphere soil of moso bamboo named JGSB02 and JGSB06.They had strong phosphorus-solubilizing capacity,and their phosphorus-dissolving capacities were 165.42 and 160.29 mg/L,respectively,their K-releasing capacities were 3.83 and 3.13 mg/L,respectively,their IAA-producing capacities were 4.38 and 10.27 mg/L respectively.JGSB02 and JGSB06 were identified as Burkholderia cepacia through the analysis of morphological characteristics,biochemical characteristics and 16S rDNA gene sequences.JGSB02 and JGSB06 had significant growth-promoting effect on the bamboo seedlings;the two strains also had antagonistic effects on the pathogenic fungi of bamboo-blight disease,with the inhibition rate of 56.67%and 66.99%respectively.The bacteria could be used as excellent biocontrol and plant growth promoting strains to promote the growth for moso bamboo,which had potential for development and application as PGPR strains.
