竹肉球内生菌多肽的分离及抑菌活性 被引量:2

摘要 采用浸提、离心、沉淀、冻干的方法得到竹肉球内生菌(Engleromyces goetzei P.Hennings)的粗提物,再经分子筛G-25凝胶柱纯化后得到粗多肽,采用滤纸片法和平板对峙法,以抑菌圈直径为指标进行抑菌活性检测,结果表明:粗多肽对农业致病真菌的菌丝生长均有不同程度的抑制作用,其中,内生菌多肽DMSO提取液对小麦赤霉病菌(Gibberella zeae)、核果炭疽病菌(Drupe anthrax)和茄子黄萎病菌(Verticilllium albo-atrum)的抑菌活性最高,IC50分别为8.70、9.48和8.96μg/mL。同时,对抗菌肽在酸碱、高温、酶解和有机溶剂环境下的稳定性进行研究,结果显示:除了具有一定的溶剂选择性及α-淀粉酶、胃蛋白酶对其有一定程度的水解之外,该抗菌肽具有很强的稳定性。用HPLC对粗多肽进一步纯化,得到电泳纯的抗菌肽。 The crude extracts of Engleromyces goetzei P.Hennings were obtained by leaching,centrifugation,precipitation and lyophilization.The crude polypeptides were obtained after extracting and isolating the crude extracts with G-25 gel chromatography.The antibacterial activities were measured with paper filtering method and plate confrontation method on the diameter of inhibition zone as an indicator.It inhibited all types of tested agricultural pathogenic fungi on mycelium growth.Among them,the antibacterial activity against Gibberella zeae,Drupe anthrax and Verticilllium albo-atrum was highest,with the IC 50 of 8.70μg/mL,9.48μg/mL and 8.96μg/mL,respectively.The crude polypeptides were further purified with HPLC to obtain an electrophoretically pure antimicrobial peptide.It had wide antimicrobial activities and was worth for the further studying.
