摘要
文章以孵化14d的鸡胚为原料,采用缓冲液法对鸡胚中的蛋白质进行提取研究,评价了其体外抗氧化活性并分析了其氨基酸组成。采用乙醚低温回流脱去鸡胚干粉中脂肪,通过差示扫描量热仪(differential scanning calorimetry,DSC)确定粗蛋白的变性起始温度为62.47℃。在单因素的基础上,应用Box-Behnken中心组合实验设计建立数学模型,得到了最优提取条件为料液比1∶16、温度50℃、时间116min,此条件下鸡胚蛋白提取率为73.87%。按此方法提取的鸡胚蛋白对1,1二苯基-2-三硝基苯肼自由基(DPPH·)、羟基自由基(·OH)具有较强的清除能力。鸡胚蛋白中必需氨基酸组成齐全,含量高,是一种理想的动物蛋白资源,有待进一步开发。
Protein extraction from fertilized egg on 14 d with a buffer solution method was studied.Amino acid composition was analyzed and the in vitro antioxidant activity of fertilized egg protein(FEP)was also evaluated.The fat was removed by diethyl ether with a method of reflux at low temperature,the denaturation temperature of the FEP was 62.47℃with differential scanning calorimetry(DSC).Based on single factor experiment,the mathematical model was established by Box-Behnken center composite design.The results showed that the optimum conditions were as follows:the ratio of solid/liquid was 1∶16,the temperature was 50℃and the time was 116 min.Under these conditions,the extraction rate of protein from fertilized egg was 73.87%.The scavenging activity against l,l-diphenyl-2-picrylhydrazyl(DPPH·)and hydroxyl radical(·OH)of the FEP was strong.FEP was rich in essential amino acid,and the amino acid composition of it was comparatively complete,which indicated that FEP was a kind of ideal animal protein resource and should be further developed.
