TASK-1对缺氧状态下人肺动脉血管环张力的影响及其机制探究

Effect of TASK-1 on the tension of pulmonary artery ring under hypoxia state and its underlying mechanism

摘要 目的 :探究串联孔域酸性敏感K通道(tandem pore domain acid-sensitive K channel,TASK-1)对缺氧状态下人离体肺动脉血管环张力的影响及其相关电生理机制。方法:手术分离并制备人离体肺动脉血管环,采用RM-6000多道生理记录仪记录给予缺氧或事先分别用PP2、PP3、bpv及A-293预孵育后再给予缺氧的血管环张力变化。原代培养人肺动脉平滑肌细胞,采用膜片钳实验分别记录缺氧、A-293、PP2、PP3、bpv处理以及用A-293、PP2、PP3、bpv预处理后再给予缺氧的TASK-1电流的变化。结果:(1)PP2预孵育后再给予缺氧处理与直接给予缺氧处理相比,人肺动脉血管环张力明显减低(P<0.05);PP3预孵育后再给予缺氧处理与直接给予缺氧处理相比,人肺动脉血管环张力差异无统计学意义(P>0.05);bpv预孵育后再给予缺氧处理与直接给予缺氧处理相比,人肺动脉血管环张力明显增加(P<0.05);A-293预孵育后再给予缺氧处理与直接给予缺氧处理相比,人肺动脉血管环张力明显减低(P<0.05)。(2)缺氧处理后人肺动脉平滑肌细胞TASK-1电流明显减少;A-293单独预处理也可明显减低TASK-1电流;A-293预孵育后再给予缺氧处理,不能进一步减低TASK-1电流;PP2预孵育后再给予缺氧处理与只用PP2处理相比,人肺动脉平滑肌细胞TASK-1电流差异无统计学意义(P>0.05);PP3预孵育后再给予缺氧处理与只用PP3处理相比,人肺动脉平滑肌细胞TASK-1电流明显减低(P<0.05);bpv预孵育后再给予缺氧处理与只用bpv处理相比,人肺动脉平滑肌细胞TASK-1电流明显减低(P<0.05)。结论:TASK-1参与缺氧后人肺动脉血管张力的调节,且此过程受c-Src的调控。 Objective:To investigate the effect of two pore domains potassium channel(TASK-1)on the tension of pulmonary artery ring under hypoxia state and its electrophysiological mechanism.Methods:Pulmonary artery rings were obtained and prepared from lung cancer patients who undergo surgery.RM-6000 multi-channel polygraph was used to record the tension of pulmonary artery rings which were directly treated with hypoxia or pre-incubated with PP2,PP3,bpv and A-293 respectively.Human pulmonary smooth muscle cells(hPASMCs)were isolated from small artery of pulmonary and cultured.Patch clamp was used to record the TASK-1 current changes of primary cultured hPASMCs which were treated with hypoxia,A-293,PP2,PP3,bpv or pre-incubated with A-293,PP2,PP3 and bpv then treated with hypoxia,respectively.Results:(1)After pre-incubated with PP2,the tension of pulmonary artery rings was significantly reduced compared with directly treated with hypoxia(P<0.05).Whether pre-incubated with PP3 or not,there was no significant difference in the tension of pulmonary artery rings(P>0.05).After pre-incubated with bpv,the tension of pulmonary artery rings was significantly increased compared with directly exposed to hypoxia(P<0.05).After pre-incubated with A-293,the tension of pulmonary artery rings was significantly reduced compared with directly exposed to hypoxia(P<0.05).(2)The TASK-1 current of human pulmonary smooth muscle cells(hPASMCs)was significantly decreased after treated with hypoxia(P<0.05).A-293 treatment also decreased the TASK-1 current of hPASMCs(P<0.05).Pre-incubated with A-293 then treated with hypoxia was not further reducing the TASK-1 current.Whether pre-incubated with PP2 or not,there was no significant difference in the TASK-1 current of hPASMCs(P>0.05).After pre-incubated with PP3,the TASK-1 current of hPASMCs was significantly decreased compared with directly treated with hypoxia(P<0.05).After pre-incubated with bpv,the TASK-1 current of hPASMCs was significantly decreased compared with directly treated with hypoxia(P<0.05).Conclusion:TASK-1 is involved in the regulation of pulmonary artery vascular tension and this process is regulated by c-Src.
